摘 要:目的揭示miR-22-3p在非小细胞肺癌(NSCLC)中的表达及作用机制。方法 q RT-PCR检测76例NSCLC组织及癌旁组织中miR-22-3p的表达,统计分析其与患者年龄、性别、肿瘤大小、病理分型、分级及淋巴结转移等临床参数的关系;生长曲线实验检测miR-22-3p对NSCLC细胞增殖的影响;在线软件Targetscan预测miR-22-3p的靶基因,荧光素酶活性分析及q RT-PCR进行验证。结果 miR-22-3p在NSCLC组织中的表达高于癌旁组织,与淋巴结转移相关,增加其在A549细胞中的表达可抑制细胞增殖;Targetscan预测到AEG-1为miR-22-3p的靶基因,荧光素酶活性分析与q RT-PCR证实miR-22-3p可与AEG-1 3′UTR结合,并可抑制A549细胞中AEG-1的表达。结论 miR-22-3p可能通过靶向AEG-1抑制NSCLC细胞增殖。Objective To investigate the expression and effect of miR?22?3p in non?small cell lung cancer(NSCLC).Methods The miR?22?3p expression level in seventy?six NSCLC tissues and para-cancer tissues was detected by qRT-PCR.The relationship between the expression of miR-22-3p and gender,age,tumor size,histology grade,pathological type and lymph node metastasis was analyzed.The function of miR-22-3p on the proliferation of NSCLC cells was tested by growth curve assay.Target genes of miR-22-3p were predicted by online software Targetscan.Luciferase reporter assay and qRT?PCR was used to certificate the prediction.Results The expression of miR-22-3p was increased in NSCLC tissues than the para-cancer tissues and was correlated to lymph node metastasis.Overexpression of miR?22?3p could suppress the proliferation of A549cells.Astrocyte?Elevated Gene-1(AEG-1)was predicted to be a target of miR-22-3p.MiR-22-3p was revealed to bind to AEG?13′UTR by luciferase reporter assay.Overexpression of miR-22-3p could inhibit the expression of AEG-1in A549cells.Suppression of miR22-3p could increase AEG-1expression.Conclusion MiR-22-3p could inhibit the proliferation of NSCLC by targeting AEG-1.
