摘  要:目的探讨银杏达莫预处理对乳鼠离体脑片谷氨酸损伤的保护机制。方法取出生7天的SD乳鼠脑皮质制备离体脑片,用2,3,5-氯化三苯基四氮唑(TTC)染色判断脑片活性后按随机法分为正常对照组(A组)、谷氨酸(Glu)损伤组(B组)、银杏达莫预处理损伤组(C组),C组于培养3天时进行药物预处理24 h(银杏达莫注射液浓度为40μg/ml);脑片培养至4天时用1 mmol/L Glu复制损伤模型,损伤30 min后将3组转入正常培养液中孵育至7天。A组不予任何处理。观察脑片细胞形态学变化,尼氏小体计数,碘化丙啶(PI)染色比例。结果 (1)脑片形态学观察:3天肉眼可见脑片轻度水肿,3~5天水肿逐渐消失,6天镜下见典型的神经细胞及少量胶质细胞;7天细胞生长最旺盛,之后凋亡逐渐占据主导。(2)苏木素-伊红(HE)染色观察细胞形态:A组神经元呈多边或梭形;B组较A组明显减少;C组神经元数量介于A组与B组之间,且组间存在差异。(3)尼氏体染色:A组尼氏小体清晰可见;B组尼氏小体阳性细胞数较A组明显减少,而C组介于A组与B组之间,(4)PI(碘化丙啶)染色:A组细胞几乎全部呈现胞核红色荧光;B组荧光明显减少、C组大量神经元出现荧光,轮廊不清。结论银杏达莫预处理对乳鼠离体脑片的Glu损伤有保护作用。Objective To study the protective mechanism of ginkgo leaf extract and dipyridamole pretreatment on glutamate injury in neonatal rat brain slices.Methods The isolated brain slices of SD rats were collected and stained with2,3,5-triphenyltetrazole(TTC)to determine the activity of brain slices.The rats were divided into normal control group(groupA),glutamate(Glu)injury group(group B),ginkgo leaf extract and dipyridamole pretreatment injury group(group C),group C was cultured for3days for24hours(ginkgo dipyrass injection concentration of40μg/Ml).Brain slices were cultured for4days and then damaged with1mmol/L Glu.After30minutes of injury,the three groups were transferred to normal culture medium for7days.Group A did not handle any treatment.To observe the morphological changes of brain cells,the number of Nisslbodies,the ratio of propidium iodide(PI)staining,Results(1)brain tablets morphological observation:3days the brain can be seen mild brain edema,3to5days of edema gradually disappeared,6days under the microscope to see the typical nerve cells and a small amount of glial cells;7days the most vigorous growth of cells,followed by apoptosis gradually dominated.(2)Hematoxylin-eosin(HE)staining was used to observe the morphological changes:the neurons in group A were polygonal or fusiform;group B was significantly lower than group A;the number of neurons in group C was between group A and group B difference.(3)Nissl body staining:group A neonatal body is clearly visible;B group of Nissl body positive cells than the A group was significantly reduced,and C group between the A group and B group,(4)PI(propidium iodide)Staining in group A showed that red fluorescence was almost all in group A;fluorescence was significantly reduced in group B,and fluorescence was observed in large groups of neurons in group C.Conclusion Ginkgo leaf extract and dipyridamole pretreatment had protective effect on Glu injury in neonatal rat brain slices.
