青蒿素对膀胱癌RT4细胞凋亡的影响

摘　　要：目的探讨青蒿素对膀胱癌RT4细胞的增殖及凋亡影响。方法选择膀胱癌RT4细胞进行体外培养，使用不同浓度（25mg/L、50mg/L、100mg/L、200mg/L)的青蒿素分别作用其24h,并运用细胞增殖CCK8法和流式细胞术（FCM)测定青蒿素对膀胱癌RT4细胞的增殖和凋亡影响。结果不同浓度的青蒿素可明显抑制膀胱癌RT4细胞生长，25mg/L、50mg/L、100mg/L和200mg/L青蒿素组的细胞生长抑制率分别为（32.00±0.13)%、（50.00±0.01)%、（65.00±0.05)%、（72.00士0.04)%,差异有统计学意义（P<0.01);其凋亡率分别为（11.69±1.97)%、（11.51±1.75)%、（12.37±2.10)%、（11.94士2.46)%,与对照组（11.92±1.14)%比较，差异无统计学意义（P>0.05);其病死率分别为（5.64±0.65)%、（9.59±1.65)%、(13.96±2.54)%、（19.68±3.63)%,与对照组（2.13±0.39)%比较，差异有统计学意义（/5<0.01)。结论青蒿素对膀胱癌RT4细胞具有明显的抑制作用，并诱导其死亡，而非促进其凋亡。Objective To investigate the effect of artemisinin on the proliferation and apoptosis of bladder cancer RT4cells.Methods Bladder cancer RT4cells were cultured in vitro and were effected for24hours with different concentrations(25mg/L^50mg/L^100mg/L^200mg/L)of artemisinin respectively.The proliferation and apoptosis of bladder cancer RT4cells were meas-ured by cell proliferation CCK8and flow cytometry(FCM).Results Different concentrations of artemisinin could significantly in-hibit the growth of bladder cancer RT4cells.The cell growth inhibitory rates of25mg/L,50mg/L,100mg/L and200mg/L arte-misinin groups were(32.00+0.13)%?(50.00+0.01)%,(65.00+0.05)%?(72.00+0.04)%,and the difference was statistically significant(P<C〇.01).Compared with the control group(11.92+1.14)%,the apoptotic rates were(11.69+1.97)%,(11.51+0.75)%,(12.37士2.10)%,(11.94士2.46)%,and the difference was not statistically significant(P>0.05).Compared with the control group(2.13+0.39)%,the mortality rates were(5.64±0.65)%,(9.59+1.65)%,(13.96±2.54)%,(19.68+3.63)%,andthe difference was statistically significant(P<C〇.01).Conclusion Artemisinin can inhibit bladder cancer RT4cells obviously and induce death,but not promote apoptosis.
