红景天苷对宫颈癌HeLa细胞增殖抑制作用的研究

摘　　要：目的:探讨不同浓度红景天苷对宫颈癌He La细胞增殖的抑制作用及其作用机制。方法:浓度分别为5,50,500μg/m L的红景天苷体外培养He La细胞24 h后,应用噻唑蓝(MTT)法观察红景天苷对He La细胞的生长抑制作用,流式细胞技术及Hoechst核染色法观察各组He La细胞凋亡情况,比色法检测凋亡相关因子Caspses-3及Caspase-9的表达。结果:红景天苷能明显抑制He La细胞增殖,不同剂量(5,50,500μg/m L)红景天苷组的细胞存活率[(82.00±3.65)%,(59.33±2.39)%,(54.00±2.61)%]与阴性对照组[(97.33±3.45)%]比较降低,差异有统计学意义(P<0.05);流式细胞技术结果显示,红景天苷低、中、高浓度组凋亡细胞比例分别为(20.53±5.76)%、(32.43±3.53)%和(42.40±3.39)%,与阴性对照组[(1.33±0.95)%]相比升高,差异均有统计学意义(P<0.01);Hoechst33258核染色法结果显示,红景天苷能够增加凋亡细胞数量;Caspase活性检测结果显示,红景天苷能增强Caspase-3及Caspase-9表达。结论:红景天苷通过增强Caspase-3及Caspase-9表达抑制宫颈癌He La细胞增殖,诱导其凋亡,提示红景天苷可能在宫颈癌的治疗中具有一定作用。Objective:To discuss different concentrations of salidroside on inhibition of cervical cancer HeLa cells proliferation and its mechanism.Methods:Concentration of5,50,500μg/mL salidroside cultured in vitro after24h,were determined by MTT method to observe for growth inhibition,flow cytometry technique and Hoechst staining were used to observe apoptosis of HeLa cells,and colorimetry was used to detect the expression of apoptosis related factor Caspase-3and Caspase-9.Results:The survival rate of different dose of salidroside were(82.00±3.65)%,(59.33±2.39)%,and(54.00±2.61)%,the difference was statistically significant(P<0.05)compared with negative control group(97.33±3.45)%;Flow cytometry technology results showed that the percentage of apoptosis cells in low,medium and high concentration salidroside group were(20.53±5.76)%,(32.43±3.53)%and(42.40±3.39)%respectively,the difference was statistically significant(P<0.05)compared with negative control group(1.33±0.95)%;Hoechst33258nuclear staining method showed that salidroside could increase the number of apoptosis cells,in addition,salidroside could also increase expression level of Caspase-3and Caspase-9.Conclusions:Salidroside inhibited cervical cancer HeLa proliferation by enhancing the expression of Caspase-3and Caspase-9,inducing its apoptosis,which indicated that salidroside may play a certain role in the treatment of cervical cancer.
