摘  要:目的研究缺氧诱导因子-1α(HIF-1α)和血管内皮生长因子(VEGF)在烟熏大鼠心肌组织中的表达,探讨吸烟对大鼠心肌损伤的影响及可能机制。方法 SD大鼠随机分入对照组和实验组,实验组给予烟熏干预,8周后观察心肌形态学改变,并通过免疫组化染色及蛋白质印迹法观察HIF-1α、VEGF在两组大鼠心肌组织中的表达情况。结果与对照组比较,实验组大鼠心肌细胞排列紊乱,且较为稀疏,部分肌原纤维扭曲甚至断裂。免疫组化染色及蛋白质印迹法结果显示,与对照组比较,实验组HIF-1α表达显著增加,而实验组VEGF表达显著下调(P<0.05)。结论吸烟可通过诱导大鼠心肌组织中HIF-1α表达上调、抑制VEGF表达进而介导心肌损伤。Objective To explore the expression of hypoxia-inducible factor-1αand vascular endothelial growth factor in cardiac muscle tissue of smoked rats,and to discuss the effect of smoking on muscle tissue of rats and possible mechanism.Methods SD rats were randomly divided into control group and experimental group,and experimental group was intervened by smoking.After8weeks,cardiac muscle tissue of rats was observed in morphology,and the expression of HIF-1αand VEGF were explored through immunohistochemical technique and Western Blot.Results Compared with control group,cardiac muscle tissue of smoked rats was arranged in disorder and sparsely,and some muscle fibril was twisty and even fractured.The expression of HIF-1αin experimental group was higher than it in control group,but the expression of VEGF in experimental group was lower than it in control group.Conclusion Smoking may damage cardiac muscle tissue of rats through increasing the expression of HIF-1αand lowering the expression of VEGF.
