摘  要:目的探讨电针百会、神庭穴对脑缺血再灌注(MCAO/R)大鼠学习记忆的影响及其可能的作用机制。方法将90只健康雄性Sprague-Dawley大鼠随机分为假手术组(n=18)和手术组(n=72)。手术组参照线栓法制备MCAO/R大鼠模型。将符合纳入标准的54只手术组大鼠随机分为模型组(n=18)、电针组(n=18)和非穴组(n=18)。电针组电针百会、神庭穴共14 d。小动物磁共振扫描检测脑梗死体积;Morris水迷宫测试检测学习记忆能力;免疫组化检测5-羟色胺1A(5-HT_(1A))受体的定位和表达。结果干预前各组大鼠脑梗死体积无显著性差异(F=1.678,P>0.05)。干预后,与模型组和非穴组相比,电针组脑梗死体积显著减小(P<0.001)。Morris水迷宫测试中,电针组较模型组和非穴组逃避潜伏期显著缩短(P<0.001),穿越平台次数增加(P<0.05)。5-HT_(1A)受体在缺血侧海马CA1、CA3和DG区均出现表达,电针组表达量较模型组和非穴组均降低(P<0.05)。非穴组各项指标与模型组比较均无显著性差异(P>0.05)。结论电针百会、神庭穴可有效改善MCAO/R大鼠的学习记忆能力,其机制可能与调节海马区5-HT_(1A)受体表达有关。Objective To explore the effect of electroacupuncture at Baihui(DU20)and Shenting(DU24)acupoints on learning andmemory ability in rats with middle cerebral artery occlusion and reperfusion(MCAO/R),and the potential mechanisms.Methods A total of90healthy male Sprague-Dawley rats were randomly divided into sham group(n=18)and operation group(n=72).The MCAO/R model wasestablished by suture method in the operation group.Finally,54qualified rats of the operation group were randomly divided into ischemiagroup(n=18),electroacupuncture group(n=18)and non-acupoint group(n=18).The electroacupuncture group received electroacupunctureat Baihui(DU20)and Shenting(DU24)for14days.The cerebral infarction volume was measured by magnetic resonance imaging(MRI).The learning and memory ability was tested by Morris water maze.The protein expression of5-hydroxytryptamine1A(5-HT1A)receptorwas detected by immunohistochemistry.Results There was no significant difference in the cerebral infarction volume among three groupsbefore intervention(F=1.678,P>0.05).Compared with the ischemia group and the non-acupoint group,the cerebral infarction volume significantlyreduced(P<0.001);the latency significantly shortened(P<0.001)and the times crossing the platform decreased(P<0.05);the expressionof5-HT1A receptor decreased in the left hippocampus(P<0.05)in the electroacupuncture group after intervention.There was no significantdifference in all the indices between the non-acupoint group and the ischemia group after intervention(P>0.05).Conclusion Electroacu-puncture at Baihui(DU20)and Shenting(DU24)could effectively increase the learning and memory ability of MCAO/R rats,which mightrelate with inhibiting the expression of5-HT1A receptor in hippocampus.
