摘  要:目的研究鼻咽癌组织中Ezrin、结肠癌转移相关基因1(metastasis-associated in colon cancer1,MACC1)及E-cadherin蛋白的表达情况,及其与放疗敏感性和预后的相关性。方法选取青岛市市立医院2011年1月至2014年1月收治的87例鼻咽癌患者作为研究对象。采用免疫组化法检测患者Ezrin、MACC1及E-cadherin蛋白的表达,分析Ezrin、MACC1及E-cadherin蛋白的表达情况与临床指标、放疗敏感性及预后的关系。结果鼻咽癌组织Ezrin、MACC1及E-cadherin蛋白的表达情况与患者年龄、性别无显著相关性(P>0.05),而与肿瘤的临床分期、浸润深度、淋巴结转移、组织分型及分化类型显著相关(P<0.05)。鼻咽癌组织Ezrin、MACC1蛋白阳性表达的放疗敏感性显著高于阴性表达(P<0.05);E-cadherin阴性表达的放疗敏感性显著高于阳性表达(P<0.05)。三者联合预测鼻咽癌放疗敏感性的有效性高于单一指标。Ezrin和MACC1蛋白阳性表达患者的3年生存率显著低于阴性表达者(P<0.05),E-cadherin蛋白阴性表达患者的3年生存率显著低于阳性表达者(P<0.05)。结论鼻咽癌组织中Ezrin、MACC1及E-cadherin蛋白的表达情况与鼻咽癌的浸润、转移有关,不仅可以了解患者病情,还可以评估患者的预后,Ezrin、MACC1、E-cadherin蛋白的表达有望成为鼻咽癌治疗的新靶点,为治疗鼻咽癌提供新的治疗策略。Objective To explore the expression of Ezrin,MACC1and E-cadherin in nasopharyngeal carcinoma and its correlation with radiosensitivity and prognosis.Method From January2012to January2014,87cases of nasopharyngeal carcinoma patients was studied in Qingdao Municipal Hospital.The expression of Ezrin,MACC1and E-cadherin were detected by immunohistochemistry,and the relationship between the expression of Ezrin,MACC1,E-cadherin and the radiosensitivity and prognosis were analyzed.Result The expression of Ezrin,MACC1and E-cadherin protein in cancer tissues were not significantly correlated with age and sex(P>0.05),and were significantly correlated with the clinical stage,depth of invasion,lymph node metastasis,histological type and differentiation type(P<0.05).The positive expression of Ezrin and MACC1protein in cancer tissues were significantly higher than that in negative expression(P<0.05);the radiosensitivity of E-cadherin negative expression was significantly higher than that in positive expression(P<0.05).The combination of the three methods was more effective than single index in predicting radiosensitivity of nasopharyngeal carcinoma.The3years survival rate of patients with positive expression of Ezrin and MACC1protein was significantly lower than that of negative expression(P<0.05),and the3years survival rate ofpatients with E-cadherin negative expression was significantly lower than that of positive expression(P<0.05).Conclusion The expression of Ezrin,MACC1and nasopharyngeal carcinoma and E-cadherin protein in nasopharyngeal carcinoma invasion and metastasis,can not only understand the patient's condition,and to assess the prognosis of patients,and the expression of Ezrin,MACC1and E-cadherin protein may be a new target for the treatment of nasopharyngeal carcinoma,providing new therapeutic strategies for the treatment of nasopharyngeal carcinoma.
