摘 要:目的研究良性胆管瘢痕CD90、α-SMA的表达.方法免疫组化SP法检测14例良性胆管瘢痕组织和10例正常胆管组织中CD90、α-SMA、Ⅰ型和Ⅲ型胶原的表达,并进行图像分析.取由良性胆管瘢痕组织培养成纤维细胞,细胞免疫荧光、流式细胞仪检测检测CD90的表达.结果 CD90、α-SMA在良性胆管瘢痕组织中表达高于正常组(P<0.05)且两者表达正相关(r=0.681,P=0.000);Ⅰ型和Ⅲ型胶原在良性胆管瘢痕组织中表达高于正常组(P<0.05)且两者表达负相关(r=-0.413,P=0.011).取由良性胆管瘢痕组织培养成纤维细胞,细胞免疫荧光、流式细胞仪检测CD90高表达.在良性胆管瘢痕组织中,CD90、α-SMA、Ⅰ型胶原、Ⅲ型胶原表达较正常组为高;CD90与α-SMA表达正相关;Ⅰ型胶原与Ⅲ型胶原表达负相关.结论良性胆管瘢痕成纤维细胞CD90高表达;CD90可能是良性胆管瘢痕异常增生的特异表型之一.Objective To investigate the expression and significance of CD90andα-SMA in benign bile ductscar.Methods Immunohistochemical staining of SP was applied to investigate the expression of protein CD90,α-SMA,type I collagen,type III collagen in14cases of benign bile duct scar and10cases of normal bile ductgroup.Finally image analysis was performed.Fibroblasts from the benign bile duct scar were cultivated.Theexpression of CD90was measured in fibroblasts by immunofluorescence and flow cytometry.Results Theexpression of CD90andα-SMA in benign bile duct scar was higher than that in normal bile duct group(P<0.05),There was positive correlation between the expression of both two in benign bile duct scar and normal bile(r=0.681,P=0.000).The expression of typeⅠcollagen and type III collagen in benign bile duct scar washigher than that in normal bile duct group(P<0.05).There was negative correlation between the expression of bothtwo in benign bile duct scar and normal bile(r=-0.413P=0.011).Fibroblasts from the benign bile duct scarwere cultivated and the excessive exprerssion of CD90was found by immunofluorescence and flow cytometry.Theexpression of CD90,α-SMA,type I collagen and type III collagen were higher in benign bile duct scar thannormal bile,In benign bile duct scar,the expression of CD90andα-SMA was positively related,type I collagenand type III collagen was negatively related.Conclusions CD90in fibroblasts of human benign bile duct scar isoverexpressed.CD90may be the specific phenotype of abnormal hyperplasia in benign bile duct scar.
