灯盏乙素对乳腺癌细胞中细胞周期调控分子的基因表达

摘　　要：目的探索灯盏乙素对肿瘤细胞周期的影响,进一步阐述灯盏乙素对肿瘤细胞的增殖抑制的作用机制.方法采用实时荧光定量PCR的方法,检测不同浓度下灯盏乙素对乳腺癌MDA-MB-231细胞周期中最重要检查点的相关因子CDK1、CDK2、cyclin A2、cyclin E2和p21的m RNA相对表达量.结果灯盏乙素的剂量低于25μmol/L时,CDK1 m RNA的相对表达量显著增加,趋势随着剂量增加而降低;CDK2 m RNA在低剂量下相对表达量显著降低,在100μmol/L浓度下表达量显著增加;cyclin A2在剂量低于50μmol/L时m RNA的相对表达量显著增加;cyclin E_2的m RNA的相对表达量在低剂量时相对表达量显著增加;p21的m RNA相对表达量在灯盏乙素剂量为50μmol/L、100μmol/L时显著增加.结论灯盏乙素显著影响CDK1、CDK2、cyclin A2、cyclin E2和p21的m RNA相对表达量,并且与剂量相关.Objective To explore the effects of scutellarin on the cell cycle,so as to demonstrate themechanism of action of scutellarin in inhibiting the proliferation of tumor cells.Methods We analyzed the relativemRNA expression of cyclin dependent kinase(CDK)1,CDK2,cyclin A2,cyclin E2and p21in a breastcancer cell line,MDA-MB-231under several doses of scutellarin by quantitative real-time PCR.The mostimportant control points were G2/Mand G1/S.CDK1and cyclin A2were related to G2/M,while CDK2and cyclinE2were related to G1/S.Results When the dose of scutellarin was less than25umol/L,the relative mRNA expressionof CDK1increased apparently.The increasing trend decreased when the doses increased.The relative mRNAexpression of CDK1hardly changed when the dose reached50μmol/L.The relative mRNA expression of cyclinA2,which regulated the G2/M phase of MDA-MB-231cells in company with CDK1,increased apparently at thedoses less than50umol/L.The relative mRNA expression of CDK2decreased apparently at low doses of scutellarin,but increased apparently at low doses of100umol/L.The relative mRNA expression of cyclin E2,which regulated the G1/S phase of MDA-MB-231cells in company with CDK2,increased apparently at low doses and hardlychanged at the dose of100μmol·L-1.The relative mRNA expression of p21,a cyclin dependent-kinaseinhibitor,increased apparently at the dose of50umol·L-1and100umol·L-1.Conclusion Scutellarin apparentlymodifies CDK1,CDK2,cyclin A2,cyclin E2and p21in a dose-dependent manner.
