绵羊红细胞与猪红细胞制备溶血素在医学免疫学实验教学中的应用比较

摘　　要：目的检测2种动物红细胞(绵羊红细胞SRBC和猪红细胞PRBC)免疫家兔所制得溶血素的效价,比较其在医学免疫学实验中的应用效果,以寻求制备符合免疫学实验及教学需求的高效价溶血素的较好方法.方法分别选择绵羊红细胞(SRBC)和猪红细胞(PRBC)经皮内多点注射、耳缘静脉注射家兔以制备溶血素,再用补体溶血试验检测2种溶血素效价的高低,并比较二者在免疫学溶血实验及教学中的运用效果.结果经补体溶血试验测得2种溶血素效价分别是:抗-SRBC为1:4 800;抗-PRBC为1:1 200.结论抗-PRBC溶血素效价低于抗-SRBC溶血素,但2种溶血素在溶血反应中均可产生较好的溶血效果,故PRBC能替代SRBC制备溶血素,并成功用于免疫学溶血实验及教学中.Objective To detect two hemolysins titer(Rabbit Anti-Sheep RBC Hemolysin and RabbitAnti-Pig RBC Hemolysin),and compare the application of these two hemolysins in immunological experimentalteaching,for selecting the better method of preparing high titer hemolysin for experimental teaching of medicalimmunology.Methods We immunized the rabbits by sheep red blood cell(SRBC)and porcine red blood cell(PRBC)through multi-point and auricular vein injection to prepare the hemolysin,then detected and compared twohemolysins titer by the complement hemolysis test.Result The hemolysin titer of the Rabbit Anti-SRBC was1:4800;and of the Rabbit Anti-PRBC was1:1200.Conclusions The hemolysin titer of the Rabbit Anti-PRBC islower than that of the Rabbit Anti-SRBC;but two kinds of hemolysin used in hemolytic reaction,can produce thegood hemolytic effect,so PRBC can replace SRBC as antigen,and immunize the rabbits for preparing hemolysin,which can be used in experimental teaching of medical immunology.
