摘  要:目的研究Corilagin对氧化型低密度脂蛋白(oxidized low density lipoprotein,OX-LDL)损伤的巨噬细胞中C反应蛋白(C-reactive protein,CRP)蛋白和m RNA表达的影响.方法体外培养小鼠单核细胞源性巨噬细胞,复制OX-LDL诱导损伤模型;Western Blot和RT-PCR方法分别观察Corilagin对巨噬细胞中CRP蛋白和m RNA表达的影响.结果与正常对照组比较,OX-LDL损伤模型组的CRP/β-actin和2-△△Ct值明显增加(P<0.05);与模型组比较,Corilagin 3.125~50μmol/L组的CRP/β-actin和2-△△Ct值明显减少(P<0.05).结论 Corilagin能明显下调OX-LDL损伤的巨噬细胞中CRP蛋白及m RNA的表达.Objective To investigate the effects of Corilagin on expression of C-reactive protein(CRP)inoxidized low density lipoprotein(OX-LDL)injured macrophage cells.Methods In vitro,micemonocyte-derived macrophages were prepared to duplicate the injury model caused by OX-LDL;Western blottingessay was used to determine the effect of Corilagin on CRP protein expression and Real time PCR assay was used tostudy the CRP mRNA expression,respectively.Results Compared with normal control group,the CRP/β-actinand2-ΔΔCt values of OX-LDL injured model group were markedly increased(P<0.05);compared with the modelgroup,Corilagin at3.125-50μmol/L obviously decreased the values of CRP/β-actin and2-ΔΔCt(P<0.05).Conclusion Corilagin could significantly down-regulate protein and mRNA expression of CRP in OX-LDL injuredmacrophage cells.
