COX-2,bcl-2和c-erbB-2在人脑胶质瘤中表达水平及其相关性研究

摘　　要：目的探讨脑胶质瘤肿瘤中bcl-2,环氧化酶-2(cyclooxygenase 2,COX-2),原癌基因HER-2/neu(c-erbB-2)的表达及二者与肿瘤恶性程度的关系。方法采用经病理和免疫组织化学染色证实的程度的低级别胶质瘤(WHO 0~Ⅱ级)25例,高级别胶质瘤(WHOⅢ,Ⅳ级)25例,对照组选用脑外伤患者的非肿瘤脑组织,用免疫组织化学法检测两组组织的bcl-2,COX-2,c-erbB-2的蛋白表达并采用半定量积分法进行结构判定。结果外伤非肿瘤脑组织中无bcl-2,COX-2,c-erbB-2的蛋白表达,低级别脑胶质瘤的bcl-2 32%,COX-2 18%,c-erbB-2 4%。高级别胶质瘤bcl-2 76%,COX-2 72%,c-erbB-2 16%,在对3组间行两两比较,bcl-2,COX-2表达蛋白表达阳性率差异具有统计学意义(P<0.05),c-erbB-2差异无统计学意义。相关分析显示,COX-2与bcl-2之间存在正相关性(r=0.682,P<0.001),COX-2与cerbB-2之间有正相关性(r=0.33,P=0.018<0.05)。bcl-2与c-erbB-2之间有正相关性(r=0.308,P=0.03<0.05)。结论 bcl-2,COX-2,c-erbB-2可能通过相关联系,影响胶质瘤的生长甚至恶变。Objective To explore the relationship between the bcl-2,COX-2,c-erbB-2protein expression and degree of malignant of human brain-glioma.Methods Useing the immunohistochemical technique,bcl-2,COX-2,c-erbB-2protein expression positive rate were determined in10no-tumor brain-tissue from brain-trauma patient who were executed the craniotomy decompression and25human low-degree(WHO0-Ⅱ)and25hunman high-degre brain-glioma(WHOⅢ,Ⅳ)specimens of different malignant degresss confirm by pathology and immunohistochemistry separatively.Results bcl-2,COX-2,c-erbB-2protein expression were not found in normal brain-tissue.the difference of bcl-2,COX-2protein expressin positive rate were statistically significant in different group,but the c-erbB-2protien expressin was no significant in three group.The relative analysis show the blc-2were positively with the expressison rate of COX-2(r=0.682,P<0.001);bcl-2were positively with the expressison rate of c-erbB-2(r=0.308,P=0.03<0.05);COX-2were positively with the expressison rate of c-erbB-2(r=0.33,P=0.018<0.05).Conclusion COX-2,bcl-2,c-erbB-2were positively relative with each other,these factors were iinfluencee the malignant degree of Brian-glioma.
