摘 要:目的研究西洋参茎叶皂苷(Panax quinquefolium saponins,PQS)对实验性糖尿病大鼠氧化损伤及其对血管内皮功能的保护作用。方法♂Wistar大鼠适应性饲养3 d后,随机抽取56只作为正常对照组,其余均采用链脲佐菌素(STZ,30 mg·kg^(-1))诱导糖尿病大鼠模型。糖尿病大鼠随机分为模型组、PQS组(100 mg·kg^(-1))和Vitamin E组(100μmol·kg^(-1)),正常组注射等量柠檬酸盐缓冲液,各组按方案给药4、8、16周后,检测大鼠血糖和血清、心、肾组织中LPO含量及SOD的活性,并记录各组大鼠胸主动脉离体血管环内皮依赖性舒张反应情况。结果与模型组比较,随着PQS给药时间的延长,PQS组大鼠的血糖及血清、心、肾组织中的LPO含量明显下降,血清、心、肾组织中SOD活性明显升高(P<0.05,P<0.01),并且随着PQS给药时间的延长,PQS组大鼠胸主动脉内皮依赖性舒张反应逐渐趋近正常值。结论PQS能通过提高机体抗氧化功能来改善糖尿病大鼠血管内皮依赖性舒张功能,从而干预糖尿病血管并发症的发生和发展。AimTo investigate the protective effect of Panax quinquefolium saponins(PQS)on oxidative damage and function of vascular endothelium.MethodsAfter3days of adaptive feeding of Wistar male rats,56rats were randomly selected as the control group,while others were induced by streptozotocin(STZ,30mg·kg-1).The diabetic rats were randomly divided into three groups:the model group,PQS group(100mg·kg-1)and Vitamin E group(100μmol·kg-1).The normal group was injected with equal amount of citrate buffer.PQS and Vitamin E groups of diabetic rats were administered orally once a day.After4,8,16weeks of administration,the concentration of blood glucose and LPO,SOD of serum,heart and kidney were measured,and the tension of the aortic vascular ring was determined.ResultsCompared with model group,with prolongation of medication,the concentration of glucose and the LPO of serum,heart and kidney in the PQS group significantly decreased(P<005,P<001),then the results of the aortic vascular ring tension showed that acetylcholine induced vasodilatation and maximum diastolic percent were obviously elevated in PQS group(P<005,P<001).ConclusionPQS could elevate the antioxidant function in diabetic rats,thus improving the endothelium dependent vasodilation function and interfering with the occurrence and development of diabetic vascular complications
