摘 要:目的:研究脑膜瘤组织中Claudins6(CLDN6)基因表达量与基质金属蛋白酶(MMPs)/基质金属蛋白酶组织抑制因子(TIMPs)、上皮间质转化(EMT)基因表达量的相关性。方法:选择在本院手术切除的脑膜瘤组织样本,同期在宜宾市第一人民医院开颅减压术中收集的正常蛛网膜组织,检测组织中CLDN6、MMPs/TIMPs、EMT基因的表达量。结果:脑膜瘤组织中CLDN6的蛋白表达量显著低于正常蛛网膜组织;脑膜瘤组织中EMMPRIN、MMP2、MMP9、Vimentin、N-cadherin的蛋白表达量均显著高于正常蛛网膜组织,TIMP1、TIMP2、E-cadherin、α-catenin的蛋白表达量均显著低于正常蛛网膜组织;CLDN6高表达脑膜瘤组织中EMMPRIN、MMP2、MMP9、Vimentin、N-cadherin的蛋白表达量显著低于CLDN6低表达脑膜瘤组织,TIMP1、TIMP2、E-cadherin、α-catenin的蛋白表达量显著高于CLDN6低表达脑膜瘤组织。结论:脑膜瘤组织中CLDN6基因的低表达能够引起MMPs水解活性增强、诱导上皮间质转化,进而促进脑膜瘤的浸润性生长。Objective:To study the correlation of Claudins6(CLDN6)gene expression in meningioma tissue with the expressions of matrix metalloproteinases(MMPs)/tissue inhibitors of matrix metalloproteinase(TIMPs)and epithelial mesenchymal transition(EMT)genes.Methods:Meningioma tissue samples that were surgically removed in Yibin First People’s Hospital between April2014and May2017were selected,and normal arachnoid tissue samples that were collected from decompressive craniectomy in this hospital during the same period were selected.The expressions of CLDN6,MMPs/TIMPs and EMT genes in tissues were determined.Results:CLDN6protein expression in meningioma tissue was significantly lower than that in normal arachnoid tissue;EMMPRIN,MMP2,MMP9,Vimentin and N cadherin protein expressions in meningioma tissue were significantly higher than those in normal arachnoid tissue while TIMP1,TIMP2,E cadherin andαcatenin protein expressions were significantly lower than those in normal arachnoid tissue;EMMPRIN,MMP2,MMP9,Vimentin and N cadherin protein expressions in meningioma tissue with higher CLDN6expression were significantly lower than those in meningioma tissue with lower CLDN6expression while TIMP1,TIMP2,E cadherin andαcatenin protein expressions were significantly higher than those in meningioma tissue with lower CLDN6expression.Conclusions:Lowly expressed CLDN6gene in meningioma tissue can increase the hydrolysis activity of MMPs,induce epithelial mesenchymal transition and thus promote the invasive growth of meningioma.
