摘　　要：目的:探讨四次跨膜蛋白1(TM4SF1)在乳腺癌组织中的表达情况,阐明其临床意义并初步探索其相关分子生物学机制。方法:收集190例人乳腺癌组织、110例癌旁组织和110例正常乳腺组织。免疫组织化学法检测乳腺癌组织、癌旁组织和正常乳腺组织中TM4SF1的表达,Western blotting法检测乳腺癌组织、癌旁组织和正常乳腺组织中TM4SF1的表达水平,RT-PCR法检测乳腺癌组织、癌旁组织和正常乳腺组织中TM4SF1mRNA的表达水平,检测不同临床病理特征乳腺癌患者乳腺癌组织中TM4SF1阳性表达率。结果:乳腺癌组织中TM4SF1阳性表达率高于癌旁组织和正常乳腺组织(P<0.05),癌旁组织与正常乳腺组织中TM4SF1的阳性表达率比较差异无统计学意义(P=0.531);TM4SF1表达与患者年龄无明显关联,与肿块大小、分化程度、淋巴结转移和肿瘤分期有密切关联(P<0.05);基底样型乳腺癌组织中TM4SF1阳性表达率高于其他3种病理类型(P<0.05)。Western blotting法检测,乳腺癌组织中TM4SF1表达水平高于癌旁组织和正常乳腺组织(P<0.05),癌旁组织与正常乳腺组织中TM4SF1表达水平比较差异无统计学意义(P>0.05)。RT-PCR检测,乳腺癌组织中TM4SF1mRNA表达水平高于癌旁组织和正常乳腺组织(P<0.01);癌旁组织与正常乳腺组织中TM4SF1mRNA的表达水平比较差异无统计学意义(P>0.05)。结论:TM4SF1在乳腺癌组织中呈高表达,TM4SF1可能通过多种机制影响乳腺癌的发生发展及远处转移,提示TM4SF1可能成为乳腺癌治疗潜在的靶点。Objective:To investigate the expression of transmembrane4super family1(TM4SF1)in breast cancer tissue,and to elucidate its clinical significance and explore the related molecular biological mechanisms.Methods:A total of190cases of human breast cancer,110cases of paracancerous tissue and110cases of normal breast tissue were collected.Immunohistochemistry was used to detect the expression levels of TM4SF1mRNA in breast cancer tissue,paracancerous tissue,and normal breast tissue;Western blotting method was used to detect the expression levels of TM4SF1in breast cancer tissue,paracancerous tissue,and normal breast tissue;RT PCR method was used to detect the expression levels of TM4SF1mRNA in breast cancer tissue,paracancerous tissue,and normal breast tissue.The positive expression rates of TM4SF1in breast cancer tissue of the breast cancer patients with different clinicopathological features were detected.Results:The positive expression rate of TM4SF1in the breast cancer tissue was significantly higher than those in paracancerous tissue and normal breast tissue(P<0.05);there was no significant difference in the positive expression rates of TM4SF1between paracancerous tissue and normal breast tissue(P=0.531);the expression of TM4SF1was not correlated with age,but was closely correlated with tumor size,differentiation degree,lymph node metastasis and tumor stage(P<0.05);the positive expression rate of TM4SF1in basal like breast cancer tissue was higher than those in the other three types of tissues(P<0.05).The results of Western blotting showed that the expression level of TM4SF1in breast cancer tissue was higher than those in paracancerous tissue and normal breast tissue(P<0.05),but there was no significant difference in the expression level of TM4SF1between paracancerous tissue and normal breast tissue(P>0.05).The results of RT PCR showed that the expression level of TM4SF1mRNA in breast cancer tissue was higher than those in the paracancerous tissue and normal breast tissue(P<0.01);there was no significant d
