摘 要:目的分析细丝蛋白A(FLNa)及转化生长因子β1(TGF-β1)在乳腺浸润性导管癌组织中的表达及其临床意义。方法利用组织芯片通过免疫组织化学方法检测88例乳腺癌、30例非癌(癌旁正常)乳腺组织石蜡标本中FLNa和TGF-β1蛋白的表达。观察FLNa、TGF-β1蛋白的表达与乳腺癌临床病理特征的关系。采用Spearman等级相关分析乳腺癌组织中FLNa和TGF-β1表达的相互关系。结果 88例乳腺浸润性导管癌组织中,FLNa蛋白的阳性表达率为56.8%(50/88),TGF-β1蛋白的阳性表达率为64.8%(57/88)。肿瘤组织中FLNa和TGF-β1蛋白的表达均高于周围正常乳腺组织的表达(P<0.05)。FLNa和TGF-β1蛋白的阳性表达与患者年龄、肿瘤大小、肿瘤TNM分期及ER、PR状态无关。FLNa在腋窝淋巴结转移组中其阳性表达率高于无淋巴结转移组(P<0.05);同时TGF-β1蛋白的表达还与HER2状态相关(P<0.05);TGF-β1蛋白的表达也与淋巴结转移状态相关(P<0.05),而FLNa和TGF-β1蛋白的表达呈正相关(r=0.211,P=0.048)。结论 FLNa和TGF-β1蛋白在乳腺癌组织中处于活化状态,联合检测FLNa和TGF-β1蛋白有助于判断乳腺癌的恶性程度和生物学行为。ObjectiveTo illuminate the correlation of the expression of filamin A(FLNa)and transform growth factor beta1(TGF-β1)in breast cancer tissue samples and its significance.MethodsThe expression of FLNa and TGF-β1protein were investigated by immunohistochemical method in88cases of human breast cancer patients,30cases of normal tissues adjacent to cancer in tissues-microarry(TMA).Expression of FLNa and TGF-β1Proteins in clinicopathological features of breast cancer was analyzed.To analyze the relationship between FLNa and TGF-β1expression in breast cancer by spearman rank correlation analysis.ResultsThe level of FLNa and TGF-β1protein expression was found to be significantly higher in breast cancer tissue than that in normal tissue(P<0.05).The level of TGF-β1protein expression was found to be significantly higher in breast cancer tissue than that in normal tissue(P<0.05).The level of FLNa and TGF-β1protein expression was not correlated with age,size,ER and PR(P>0.05),However,expression of FLNa was significant higher with lymph node metastasis than that without lymph node metastasis(P<0.05).Moreover,there was a positive correlation between the expression of FLNa and TGF-β1protein in breast cancer(r=0.211,P=0.048).The expression of TGF-β1was positively correlated with lymph nodes metastasis and HER2stage(P<0.05).ConclusionFLNa and TGF-β1protein are overexpressed in breast cancer tissues,which are associated with lymph node metastasis of breast cancer.FLNa and TGF-β1protein are activated and play an important role in development of breast cancer.Effective detection of FLNa and TGF-β1proteins may contribute to the evaluation of malignancy and biological behavior of breast cancer.
