摘  要:为进一步明确导致新乡地区金银花根腐病的主要病原及其种属地位,采用纯培养技术对金银花根腐病的病原菌进行分离、纯化,并利用根部切伤接种法对病原回接以测定其致病性,通过显微观察和核糖体DNA转录间隔区(ITS)序列分析对病原菌进行鉴定.结果表明:PDA培养基上的C-3-1菌落为绒毛状,大分生孢子为镰刀形,有2~3个隔膜,大小为20.0~35.1μm×3.5~5.0μm,小分生孢子为肾形,有0~1个隔膜,大小为5.0~12.0μm×2.0~4.0μm.用通用引物ITS1和ITS2扩增真菌r DNA的内部转录间隔区ITS序列与尖孢镰刀菌C-3-1基因全序列(登录号:JF439472,GQ922558,DQ452447.1)相似性为100%.确定尖孢镰刀菌Fusarium oxysporum为新乡金银花根腐病的病原菌.In order to confirm root rot pathogen on Lonicera japonica in Xinxiang,using pure culture techniques,the Lonicera japonica root rot pathogen was isolated,and its pathogen was further assessed using a root inoculation method.The causal pathogen was identified by microscopic observation and molecular method.One root rot pathogen fungus was isolated from the infencted plants of Lonicera japonica:the colony of C-3-1on PDA media was velvet,and root-cells of big conidiospore were pointed,measured2~3estrangements and20.0~35.1μm×3.5~5.0μm;small conidiospores were reniform,measured0~1estrangements and5.0~15.0μm×2.0~4.0μm.To further confirm identity of the causal fungus,the internal transcribed spacer(ITS)of fungal rDNA was amplified with universal primers ITS1and ITS2,and sequenced.The ITS sequence shows100%similarity with that of C-3-1on Fusarium oxysporum(Accession No.JF439472,No.GQ922558,No.DQ452447.1),and they were clustered into one clade.The Lonicera japonica root rot pathogen fungus is Fusarium oxysporum.
