摘  要:[目的]比较四种染色方法制作白血病细胞血涂片标本的效果。[方法]Wright氏染色法:染色时间分别为10、15、20 min;Giemsa氏染色法:染色时间分别10、15、20 min;Wright氏-Giemsa氏混合染色法,染色时间分别10、15、20 min;Wright氏-Giemsa氏分步联合染色法,染色时间分别10、15、20 min。[结果]Wright氏-Giemsa氏分步联合染色法对前三种染色法起互补作用,其染色法制作的标本兼有前三种方法的优点,即对细胞核着色程度适中,细胞核结构和色泽清晰艳丽,对细胞核结构的识别较佳;可以使细胞质颗粒着色较好,色泽纯正,易于观察辨认。[结论]Wright氏-Giemsa氏分步联合染色法制作的标本,染色效果最佳,易于观察辨认,值得推广。〔Objective〕The effects of four kinds of staining methods on the preparation of leukemia cells were compared.〔Methods〕By Wright′s Staining,the staining time was10min,15mim,20min;2.By Giemsa′s staining method,the dyeing time was10min,15min and20min;3.By Wright Giemsa mixed staining method,the dyeing time was10min,15min and20min;4.By Wright Giemsa step by step staining method,the staining time was10min,15min and20min.〔Results〕Wright Giemsa step by step combination method was complementary to the first three staining methods.The stained samples had the advantages of the first three methods which included the proper staining of the nucleus with clear structure and color of the nucleus,better identification of the nuclear structure and better staining of cytoplasm particles and easier observation of its structure.〔Conclusion〕Wright Giemsa step by step combination of staining method produced the best results,made observation and identification easier,and therefore worth being recommended to the professionals.
