摘  要:目的观察补肾活血中药对大鼠慢性高眼压模型视网膜Bcl2及Bad的干预作用,探讨其作用机理。方法选取30只SD大鼠,随机分为对照组、给药组及模型组,每组各10只。采用烙闭巩膜上静脉方法制作慢性高眼压大鼠动物模型,每组连续灌胃8周后处死大鼠,观察补肾活血法对EIOP大鼠眼压、视网膜神经节细胞超微结构和视网膜Bcl2、Bad表达的影响。结果SD大鼠造模后即刻直至造模后8周与造模前眼压相比均升高,有统计学差异(均P<0.01),造模后8周眼压与造模后即刻相比,给药组有降低趋势,差异有统计学意义(P<0.01),模型组差异无统计学意义(P>0.05);造模后8周,给药组Bcl2阳性表达最高,与对照组和模型组相比,差异有统计学意义(均P<0.01),模型组Bcl2表达多于对照组,差异具有统计学意义(P<0.01),模型组Bad阳性表达最高,与给药组和对照组相比,差异有统计学意义(均P<0.01),给药组的Bad的表达多于对照组,差异具有统计学意义(均P<0.01)。给药组RGCs超微结构较模型组明显改善。结论补肾活血中药(杞菊地黄丸合复方丹参片)能保护SD大鼠慢性EIOP模型视功能,主要表现为:上调Bcl2的表达、下调Bad的表达、降低眼压及改善RGCs超微结构。ObjectiveTo observe the effect of traditional Chinese medicine(TCM)of BuShenHuoXue on retinal BCL2and Bad in SD rat model of chronic elevated intraocular pressure,and explore it’s initial mechanism.MethodsThe rats were randomly divided into3groups:control group,model group and treatment group,10rats in each group.The model of chronic EIOP was established by unilaterally cauterizing3episcleral vessels.After given TCM of BuShenHuoXue or normal saline for8weeks,the rats were put to death.The effect of intraocular pressure(IOP),expression of retinal BCL2and Bad,ultrastructure of RGCs were observed.ResultsCompared with pre operation,IOP in treatment group and model group at model building until postoperation8weeks were increased(all P<0.01).There was obviously reduce in treatment group between postoperation8weeks and models building(P<0.01),While model group was not statistically significant(P>0.05).8weeks after modeling,the Immunohistochemical detection of retina showed that the treatment group’s expression on Bcl2was the highest,compared with the control group and modle group,there were statistically differences(all P<0.01),model group’s expression on Bad was the highest,compared with the treatment group and control group,there were statistically differences(all P<0.01).The ultrastructure of RGCs in the treatment group was significantly improved compared with model group.ConclusionTCM of BuShenHuoXue(QijuDihuang pills and DaShen tablets)can protect the visual function on the rat model of chronic EIOP.It mainly manifested as follows:up regulated the expression of retinal Bcl2,dwonregulated the expression of retinal Bad,reduced IOP slightly and improved the ultrastructure of RGCs.
