摘 要:目的观察补精益视片对慢性高眼压模型(elevatedintraocularpressureEIOP)大鼠外侧膝状体中尼氏小体(nisslbody)含量的影响,探讨补精益视片视功能保护的作用机理。方法选取30只SD大鼠,随机分为对照组、给药组及模型组,每组各10只。采用烙闭巩膜上静脉方法制作慢性高眼压大鼠动物模型,每组连续灌胃8周后处死大鼠,观察补精益视片对EIOP大鼠眼压、外侧膝状体(lateralgeniculatenucleusLGN)尼氏小体含量的影响。结果相比造模前,从造模即刻开始至造模后8周,给药组及模型组大鼠的眼压均有明显升高,差异具有统计学意义(均P<0.01);造模后8周与造模即刻相比,给药组眼压有降低趋势,差异有统计学意义(P<0.01),而模型组眼压无降低趋势;模型组尼氏小体总面积(55863.31±18427.166S/μm2)及积分光密度(16084.76±6013.412)较对照组(114533.71±17849.482S/μm2,35237.50±4626.313)及给药组(108020.95±27719.546S/μm2,31261.33±7800.446)明显偏低,差异具有统计学意义(均P<0.01),给药组尼氏小体总面积及积分光密度较对照组稍偏低,差异无统计学意义(P>0.05)。结论补精益视片能提高慢性EIOP模型大鼠LGN尼氏小体含量,从而发挥视功能保护作用。ObjectiveTo observe the effect of BuJingYiShi tablets on lateral geniculate nucleus'(LGN)Nissl body damage in rat model of chronic elevated intraocular pressure(EIOP),and explore the mechanism of it initially.MethodsThe rats were randomly divided into3groups:control group,model group and treatment group,10rats in each group.The model of chronic EIOP was established by unilaterally cauterizing3episcleral vessels.After given BuJingYiShi tablets and normal saline for8weeks,killed the rats and taken out the brains.IOP,content of nissl bodies in lateral geniculate nucleus(LGN)were observed.ResultsImmediately modeling and8weeks after modeling,IOP were highly significant different from before modeling(all P<0.01).There was a decreasing tendency in treatment group between8weeks after modeling and immediately modeling(P<0.01).While model group was not statistically significant(P>0.05).The total area(55863.31±18427.166S/μm2)and integrated optical density(16084.76±6013.412)of Nissl body in model groups were lower than treatment group(114533.71±17849.482S/μm2,35237.50±4626.313)and control group(108020.95±27719.546S/μm2,31261.33±7800.446)(all P<0.01);Though total area and integrated optical density in treatment group were little lower than control group,there was no significant difference in statistics(P>0.05).ConclusionBuJingYiShi tablets can protect the visual function on the rat model of chronic EIOP by enhancing content of nissl bodies in LGN.
