摘 要:目的探讨差异表达miRNAs与甲状腺癌患者预后的关系。方法从肿瘤基因组图谱(TCGA)数据库下载甲状腺癌及甲状腺癌旁组织的miRNAs测序数据和临床信息。采用R程序语言包筛选差异表达的miRNAs。应用Kaplan-Meier(log-rank检验)生存曲线进行生存分析,比较与总体生存率相关的miRNAs与临床参数的关系。利用靶基因分析软件对与总体生存率相关miRNAs的靶基因进行预测,并进行基因本体(GO)功能和京都基因与基因组百科全书(KEGG)通路富集分析。结果筛选出差异表达miRNAs共计91个,其中上调miRNAs 86个,下调miRNAs 5个。miRNA-509-3、miRNA-196a-2和miRNA-767与总体生存率呈显著相关(P<0.05)。miRNA-509-3和miRNA-196a-2与临床分期和甲状腺癌类型显著相关(P<0.05)。3种miRNAs的靶基因主要参与细胞黏附、凋亡、细胞周期等生物学过程和与肿瘤相关的代谢通路。结论 miRNA-509-3、miRNA-196a-2和miRNA-767可能在甲状腺癌的发生发展中发挥作用,通过对3种miRNAs的靶基因预测分析,为甲状腺癌发病机制的进一步研究提供基础。Objective To investigate relationship between differentially expressed miRNAs and prognosis of patients with thyroid cancer.Methods The miRNAs sequencing data and clinical informations of thyroid cancer and para carcinoma tissues were downloaded from the cancer genome atlas(TCGA)database,and differentially expressed miRNAs were screened using R programming language package.Kaplan Meier(log rank test)survival curve was used for survival analysis,and relationship between miRNAs and clinical parameters associated with overall survival rate was analyzed.Target gene of miRNAs associated with overall survival rate was pretested using target gene analysis software,and gene ontology(GO)function with the Kyoto encyclopedia of genes and genomes(KEGG)pathway enrichment were analyzed.Results A total of91differentially expressed miRNAs were screened,which included86up regulated miRNAs and5down regulated miRNAs.MiRNA5093,miRNA196a2and miRNA767were significantly correlated with overall survival rate(P<0.05).The miRNA5093and miRNA196a2were significantly correlated with clinical stage and cancer type(P<0.05).The three target genes of miRNAs were mainly involved in biological process such as cell adhesion,apoptosis and cell cycle and cancer related metabolic pathways.Conclusion The miRNA5093,miRNA196a2and miRNA767may be involved in pathogenesy and development of thyroid cancer.Prediction analysis for the three target genes of miRNAs may provide base for further studying mechanism of thyroid cancer.
