摘  要:目的探讨长链非编码尿路上皮癌相关基因1(lncRNA-UCA1)调控miR-143表达影响前列腺癌细胞生物学行为的作用及机制。方法 q PCR检测lncRNA-UCA1和miR-143在前列腺癌组织和癌旁正常前列腺组织中的表达情况。双荧光素酶报告基因检测lncRNA-UCA1与miR-143的相互作用。克隆形成实验检测抑制lncRNA-UCA1表达后前列腺癌细胞增殖能力的变化。凋亡实验检测抑制lncRNA-UCA1表达后前列腺癌细胞凋亡行为的变化。结果前列腺癌组织中lncRNA-UCA1的表达水平高于正常前列腺组织(P<0.05),miR-143的表达水平低于正常前列腺组织(P<0.05)。lnRNA-UCA1+siRNA可以明显抑制miR-143-WT的荧光素酶活性(P<0.05)。lncRNA-UCA1+siRNA组细胞增殖率低于NC组(P<0.01),细胞凋亡率高于NC组(P<0.05)。结论 lncRNA-UCA1可以靶向调节miR-143调控前列腺癌细胞的增殖和凋亡行为。Objective To investigate effect and mechanism of miR-143expression regulated by long chain non coding urothelial carcinoma associated gene1(lncRNA-UCA1)on biological behavior of prostate cancer cells.Methods Quantitative polumerase chain reaction(qPCR)was used to detect lncRNA-UCA1and miR-143expressions in prostate cancer and adjacent normal prostate tissues.The double luciferase reporter gene was used to detect relationship between lncRNA-UCA1and miR-143.Clone formation experiment was used to detect changes of proliferation ability of prostate cancer cells after inhibiting lncRNA-UCA1expression.Apoptosis experiment was used to detect apoptosis changes of prostate cancer cells after inhibiting lncRNA-UCA1expression.Results In prostate cancer tissues,lncRNA-UCA1expression was significantly higher(P<0.05),while miR-143expression was significantly lower than those in normal prostate tissues(P<0.05).LnRNA-UCA1+siRNA could significantly inhibit luciferase activity of miR-143-WT(P<005).In lncRNA-UCA1+siRNA group,cell proliferation rate was significantly lower(P<0.01),while apoptosis rate was significantly higher than those in NC group(P<0.05).ConclusionlncRNA-UCA1may targeted regulate miR-143to regulate proliferation and apoptosis of prostate cancer cells.
