摘 要:目的:探讨Smo和Gli1在胃癌及癌旁组织中的表达与胃癌的临床病理因素之间的关系;并探讨两者之间在胃癌中的关系。方法:应用免疫组织化学Eli Vision法测定75例胃癌组织和45例对应的癌旁组织中Smo和Gli1的表达情况。应用RTPCR技术检测30例新鲜胃癌组织及癌旁组(距切缘5 cm)组织中Smo m RNA和Gli1 m RNA的表达情况。结果:免疫组织化学染色结果显示,胃癌组Smo和Gli1表达阳性率均显著高于癌旁组(P<0.01);RT-PCR显示,胃癌组Smo和Gli1表达水平均较癌旁组显著升高(P<0.01)。2种方法均提示Smo、Gli1在胃癌组织中的表达阳性率在病人年龄、性别、肿瘤分化程度及肿瘤大小间差异均无统计学意义(P>0.05),而在TNM分期、浸润深度、及淋巴结转移间差异均有统计学意义(P<0.01)。结论:Smo与Gli1蛋白表达与胃癌淋巴结转移、临床分期、浸润深度等因素相关,提示两者可能与胃癌的恶性生物学行为有关。Smo与Gli1在胃癌中的表达呈正相关关系,两者可能协同促进肿瘤的发生与发展。Objective:To investigate the expressions of Smo and Gli1in gastric cancer tissues and paracarcinoma tissues,and their correlations with clinicopathological features of gastric cancer.Methods:The protein expressions of Smo and Glil in75cases of gastric cancer tissues and45cases of paracarcinoma tissues were detected using immunohistochemical EliVision method.The mRNA expressions of Smo and Glil in30cases of fresh gastric cancer tissues and paracarcinoma tissues(5cm distance from cut edge)were detected using RT-PCR technology.Results:The results of immunostochemical staining showed that the positive rates of the protein expressions of Smo and Glil in gastric cancer tissues were significantly higher than those in paracarcinoma tissues(P<0.01).Compared with the paracarcinoma tissues,the results of RT-PCR showed that the mRNA expression levels of Smo and Gli1significantly increased in gastric cancer tissues(P<0.01).Both methods demonstrated that the differences of the positive expression rates of Smo and Gli1in different age,gender,tumor differentiation and size were not statistically significant(P>0.05),and the differences of the expression positive rates of Smo and Gli1in different TNM stage,invasion depth and lymph node metastasis were statistically significant(P<0.01).Conclusions:The protein expressions of Smo and Gli1are related to the lymph node metastasis,clinical stage and invasion depth of gastric cancer,which hints that Smo and Gli1may be related to the malignant biological behavior of gastric cancer.The expression of Smo is positively correlated with Glil in gastric cancer,which maybe play a synergistic action in the occurrence and development of gastric cancer.
