摘  要:目的探讨二苯乙烯苷(tetrahydroxystilbene glucoside,TSG)对沙鼠脑缺血/再灌注(ischemia/reperfusion,I/R)神经元损伤保护作用的机制。方法将42只沙鼠随机分成6组,脑I/R模型对照组(I/R)、假手术(Sham)组、TSG小剂量(1.5mg/kg)组、TSG中剂量(3mg/kg)组、TSG大剂量(6mg/kg)组以及阳性药依达拉奉对照组(3 mg/kg);6d后HE染色观察脑组织形态学改变,免疫组织化学法检测脑组织p53的表达,Western blot法检测海马CA1区Bcl-2、Bax的表达。结果与假手术组相比,I/R组脑组织中p53阳性细胞数显著增多,海马CA1区Bax水平显著增加,Bcl-2水平显著降低。而依达拉奉阳性对照组和TSG高、中剂量组均可以显著上调Bcl-2水平,降低p53及Bax水平。TSG小剂量组对p53、Bcl-2、Bax水平均没有明显作用。结论 TSG对于I/R引发脑神经元损伤保护作用的机制可能与影响p53、Bcl-2、Bax凋亡相关因子表达有关。Objective To investigate the protective effects of tetrahydroxystilbene glucoside(TSG)on ischemia-reperfusion(I/R)-induced brain injury and the underlying mechanisms in gerbils.Methods Forty two gerbils were randomly divided into6groups:I/R group,sham group,I/R+low TSG group(1.5mg/kg),I/R+medium TSG group(3mg/kg),I/R+high TSG group(6mg/kg)and I/R+edaravone control group(3mg/kg).Six days after reperfusion,the morphological changes of brain tissues were observed through hematoxylin and eosin stain and the expression of p53in brain cortex was examined by the immunohistochemistry.The expressions of Bax and Bcl-2in hippocampal CA1region were determined by Western blot.Results Compared to Sham group,the number of p53positive cells as well as Bax level in I/R group increased significantly.In contrast,Bcl-2level in hippocampal CA1region decreased dramatically.Edaravone and TSG treatment at both medium and high but not low doses evidently reversed the above changes.Conclusion TSG had protective effects on I/R induced brain injury,which may be related to its regulation on the expression of apoptosis-related factors including p53,Bax and Bcl-2.
