大蒜素对小鼠酒精性肝纤维化的预防作用研究

摘　　要：研究大蒜素对酒精性肝纤维化的预防作用。选用SPF级昆明小鼠40只,分为正常对照组、酒精性肝纤维化组、护肝片组、大蒜素高和低浓度组,各组灌服0.2 m L相应药物30 d后使用红星二锅头建立酒精性肝纤维化模型,测定肝功能相关指标,免疫印迹法检测肝组织中肿瘤坏死因子-α(TNF-α)、转化生长因子-β1(TGF-β1)和饰胶蛋白聚糖(Decorin)的蛋白表达。大蒜素能够降低小鼠ALT、AST活性和MDA、LDL-C含量,增加SOD、GSH-PX活性和GSH含量。Western blotting显示大蒜素组肝组织中TNF-α和TGF-β1的表达水平显著降低,而Decorin表达水平升高。大蒜素能够增强酒精性肝纤维化小鼠肝脏抗氧化酶活力,降低其肝脏的脂质过氧化水平,干预TNF-α、TGF-β1和Decorin等肝纤维化相关蛋白的表达,对小鼠肝脏具有一定的保护作用。To explor the preventive effects of allicin on alcohol liver fibrosis.Forty KM mice were randomly divided into normal control,alcohol liver fibrosis,low concentration and high concentration groups respectively.0.2mL corresponding drugs were given for30d.The Red Start Erguotou was given to build model.The expression of TNF-α,TGF-β1and Decorin were detected by Western blotting.ALT,AST,MDA and LDL-C decreased,SOD,GSH-PX and GSH increased with allicin administration.The expression of TNF-αand TGF-β1were inhibited,Decorin was improved significantly in liver tissue.Allicin can enhance reduce lipid peroxide,and interfere the expression of TNF-α,TGF-β1and Decorin.Allicin has certain effects on alcohol liver fibrosis in mice.
