摘 要:目的:检测泛素化特异性蛋白酶39(ubiquitin-specific protease,USP39)在宫颈癌组织中的表达,并分析USP39的表达水平与宫颈癌患者的临床病理特征及术后生存周期的相关性。方法:采用实时定量聚合酶链反应(qRT-PCR)技术检测94例宫颈癌组织及癌旁组织中USP39 mRNA表达水平,采用Spearman秩相关分析USP39与宫颈癌患者临床病理特征的相关性,绘制Kaplan-Meier生存曲线,采用Log-rank检验分析USP39高表达和低表达的宫颈癌患者5年总体生存率的差异。结果:qRT-PCR结果显示USP39 mRNA在宫颈癌组织中的表达水平显著高于癌旁组织(P<0.05)。Spearman秩相关分析结果显示USP39的表达水平与宫颈癌患者淋巴转移及FIGO分期呈正相关(r分别为0.732和0.625,P<0.05),而与患者其他临床病理特征无显著相关性。Kaplan-Meier生存曲线结果显示USP39高表达的宫颈癌患者术后5年总体生存率低于USP39低表达的宫颈癌患者(P<0.05)。结论:USP39在宫颈癌组织中表达水平升高,可作为判断宫颈癌患者生存周期的潜在分子标志物。Objective:To explore the expression of ubiquitin-specific protease 39(USP39)in cervical cancer tissues,and to investigate the association of the expression level of USP39 with clinicopathological features and postoperative survival time of cervical cancer patients.Methods:Quantitative real-time polymerase chain reactions(qRT-PCR)was conducted to analyze the expression levels of USP39 mRNA in 94 cases of cervical cancer tissues and tumor-adjacent tissue.Spearman rank correlation and Kaplan-Meier survival curves were conducted to analyze the clinical correlation of USP39 expression with 5-year overall survival rate.Results:qRTPCR results revealed that the expression level of USP39 was upregulated in cervical cancer tissues compared with tumor-adjacent tissues(P<0.05).Spearman rank correlation showed that high USP39 expression was significantly associated with lymph node metastosis and FIGO stage of cervical cancer patients(r=0.732,0.625,P<0.05).Kaplan-Meier survival curves showed that high USP39 expression was negatively correlated with 5-year survival overall rate(P<0.05).Conclusion:USP39 expression is upregulated in cervical cancer tissues,which can be a potential survival biomarker in cervical cancer.
