摘 要:探讨胰高血糖素(Glu)在适量饮酒小鼠及1型糖尿病(T1DM)饮酒小鼠发病过程中的可能作用。采用正常雄性C57BL/6J小鼠通过灌胃饮用54%vol董酒及注射链脲佐菌素建立单纯饮酒、糖尿病、糖尿病饮酒模型,测空腹血糖,通过免疫组织化学、real-time PCR,图像分析及形态计量法观察胰岛α细胞分泌Glu及胰腺Glu m RNA表达的改变。结果表明,(1)糖尿病组及糖尿病饮酒组小鼠空腹血糖升高,达到小鼠糖尿病成模标准;(2)单纯饮酒组小鼠α细胞表达Glu平均光密度、面数密度无明显变化;(3)糖尿病组及糖尿病饮酒组α细胞平均光密度值无明显升高,而面数密度与Glu mRNA相对表达量逐渐增高(P<0.05)。结果显示,在本实验设定的剂量及时长内,单纯饮酒小鼠Glu表达改变不明显;适量饮用该白酒对糖尿病小鼠Glu表达未见明显影响。The aim of this study was to investigateαcell population and secretion in mice with T1DM by moderate intragastric administrationof Dongjiu.Male C57BL/6J mice were divided into3groups through intragastric administration of54%vol Dongjiu andinjection with STZ:liquor consumption group,diabetes group and diabetes-liquor consumption group.Fasting blood-glucose was measured and Glu mRNA expression change in pancreas was assessed by immunohistochemical SABC,real-time PCR,image analysis and morphmetric methods.The results suggested that,fasting blood glucose increased above11.1mmol/L in diabetes group and diabetes-liquor consumption group,which met the modeling standards of mice with T1DM;there was no significant change in the values of mean density and numerical density on area(NA)of Glu mRNA in liquor consumption group;the values of mean density of Glu mRNA kept steady in diabetes group and in diabetes-liquor consumption group,but NA and Glu mRNA expression kept increasing gradually(P<0.05).The findings revealed that,within the designed period and dosage in our experiment,there was no evident change in Glu expression in liquor consumption group,and moderate liquor intake had no significant effects on Glu expression in mice with diabetes.
