摘  要:目的建立及优化骨髓活检石蜡切片原位荧光杂交(FISH)检测流程。方法建立和优化骨髓活检石蜡切片FISH检测步骤和实验条件,在20例骨髓活检组织,其中5例诊断为套细胞淋巴瘤(MCL),应用FISH检测IGH/CCND1探针。结果建立骨髓活检石蜡切片的FISH检测步骤和实验条件,5例确诊MCL骨髓活检石蜡切片可检测到IGH/CCND1融合信号。证明本方法的可行性。结论骨髓活检石蜡切片FISH检测可以获得信号清晰、核形态较好、背景清晰的FISH染色结果,可以满足临床应用要求。Objective To establish and optimize the process of fluorescence in situ hybridization(FISH)for paraffin sections of bone marrow biopsies.Methods The testing step and experimental conditions of FISH in paraffin sections of bone marrow biopsies,and 20 cases of bone marrow biopsies including 5 cases diagnosed as mantle cell lymphoma(MCL),and IGH/CCND1 fusion gene was detected by FISH in paraffin sections.Results The experimental methods and conditions of FISH in paraffin sections of bone marrow biopsies were successfully established and these all 5 MCL cases were successfully foudd that they had IGH/CCND1 fusion gene which convinced the feasibility of this method.Conclusion The establishment of operating process of FISH in paraffin sections of bone marrow biopsies can achieve a good result with clean signal,good nuclear shape and clean background,thus it can meet the clinical demands.
