摘  要:目的研究盐酸二氧丙嗪(DPZ)与DNA间的相互作用。方法采用光谱法、盐效应作用及热力学理论对盐酸二氧丙嗪与DNA间相互作用进行研究。结果随着温度升高,结合常数Ka下降;热力学参数ΔH0<0,ΔS0>0;氯化钠和磷酸二氢钠的加入对盐酸二氧丙嗪-DNA体系的荧光强度影响不大,盐酸二氧丙嗪的加入可使溴化乙锭-DNA体系的荧光强度增强。结论盐酸二氧丙嗪以静态淬灭形式与DNA发生相互作用,二者间作用力主要为氢键和疏水作用力,盐酸二氧丙嗪与DNA分子间结合模式为与溴化乙锭类似的嵌插模式。Objective To study the interaction between dioxoporphyrin hydrochloride(DPZ)and DNA.Methods Spectroscopic methods,salt effect and thernodynamics theor were adopted in this paper.Results The binding constant(K,a)of DPZ with DNA decreased with the increase of temperature.The thernodynamic parameters of the interaction by a double reciprocal method were calculated:AH0<0,AS0>0.The addition of NaCl and NaH2P04had no effect on the fluorescence intensity of DPZ-DNA system.The addition of DPZ had effect obviously on the fluorescence intensity of EB-DNA system.Conclusion The quenching mechanism between DPZ and DNA was static quenching.They were bounded mostly by hydrogen bonding and hydrophobic interaction force.The binding mode of DPZ binding to DNA was similar to EB,which was insert mode.
