摘　　要：目的探讨miR-181-5p通过靶向作用AKT3通路,从而影响肝癌细胞增殖、侵袭和转移。方法利用q RT-PCR技术检测人肝癌标本与正常肝组织中miR-181-5p、AKT3含量,并且采用免疫组化染色检测miR-181-5p高表达肝癌组织和miR-181-5p低表达的肝癌组织中AKT3、E-cadherin和N-cadherin的含量及分布情况;体外培养肝癌细胞SMMC-7721,采用miR-181-5p mimic或inhibitor处理以促进或抑制肝癌细胞miR-181-5p表达,AKT3si RNA沉默AKT3基因;平板克隆实验和Transwell实验检测肝癌细胞增殖、侵袭和转移能力。结果肝癌组织中miR-181-5p呈低表达水平,而AKT3表达升高。miR-181-5p高表达肝癌组织较miR-181-5p低表达的肝癌组织中AKT3、E-cadherin和N-cadherin的表达量降低。通过si RNA抑制AKT3表达,可以抑制肝癌细胞的增殖、侵袭和转移。结论 miR-181-5p在肝癌组织中低表达,miR-181-5p能抑制AKT3表达,从而抑制肝癌细胞增殖、侵袭和转移。Objective To investigate the effect of miR-181-5p on the proliferation,invasion and metastasis of liver cancer cells by targeting AKT3 pathway. Methods The content of miR-181-5p and AKT3 in human hepatocellular carcinoma and normal liver tissues were detected by qRT-PCR.The contents and distributions of AKT3,E-cadherin and N-cadherin in miR-181-5p low-expressed and high-expressed hepatocellular carcinoma tissues were detected by immunohistochemical staining.Liver cancer cells SMMC-7721 were cultured in vitro.miR-181-5p mimic or inhibitor were taken to promote or inhibit the expression of miR-181-5p in liver cancer cells.AKT3 siRNA silenced AKT3-related genes.Platelet cloning assay and transwell assay were carried out to detect the proliferation,invasion and metastasis of hepatocarcinoma cells.Results The expression of miR-181-5p was down-regulated expression in HCC tissues,while the expressions of AKT3 were up-regulated expression.The expression of AKT3,E-cadherin and N-cadherin in miR-181-5p high-expressed hepatocellular carcinoma was lower than that in miR-181-5p low-expressed hepatocellular carcinoma.Inhibition of AKT3 expression by siRNA could inhibit the proliferation,invasion and metastasis of liver cancer cells. Conclusion miR-181-5p can inhibit AKT3,thus it can inhibit the proliferation,invasion and metastasis of hepatocarcinoma cells.
