摘  要:为探讨Iss融合蛋白在鸡大肠杆菌病中的免疫保护作用,试验采用GST-Iss融合蛋白免疫14日龄雏鸡,通过动物体内攻毒试验及体外血清抗性试验,分析Iss抗血清的免疫保护作用。间接ELISA检测血清抗体效价,间接ELISA、凝集试验检测抗血清对鸡致病性大肠埃希菌HO2、CVCC1553菌株的吸附水平。通过iss基因、ybtA基因、chuA基因实时荧光定量PCR,检测2株菌株的mRNA转录水平。结果发现,Iss融合蛋白免疫雏鸡,在体外试验中能够抑制HO2菌株的血清抗性,并抑制其iss基因、ybtA基因及chuA基因的转录,但对CVCC1553菌株无抑制作用。体内试验未产生明显保护作用。因此,单一毒力因子制备的亚单位疫苗的免疫效果有限,探讨多基因联合的亚单位疫苗具有更好的实际意义。To explore the immune effect of iss protein in chicken colibacillosis,14-day-old chickens were immunized with GST-Iss fusion protein,followed by the infection test and serum resistance test(in vitro).Indirect ELISA and agglutination test were used to detect the adsorption level of serum with pathogenic Escherichia coli HO2 and CVCC1553 strains.By real-time fluorescence quantitative PCR tests,the transcriptional levels of iss gene,ybtA gene and chuA gene were detected.The results showed that the Iss fusion protein had a protective effect on HO2 stain in vitro,and the antiserum was able to inhibit the serum resistance of HO2 strains by inhibiting the transcription of the genes,except CVCC1553.But there was no significant protective effect in vivo test.Therefore,the immune effect of subunit vaccine developed from single virulence factor is limitied,the investigation the potential of multi-gene subunit vaccine may be useful.
