摘 要:目的建立桑枝中氨基酸成分的薄层鉴别方法。方法以蒸馏水为提取溶剂对桑枝中氨基酸进行超声提取,并对展开系统[正丁醇-冰醋酸-水(4∶2.2∶3)、正丁醇-冰醋酸-水(4∶1∶5)、正丁醇-冰醋酸-水(4∶3∶1),正丁醇-冰醋酸-水(4∶2.2∶1),正丁醇-冰醋酸-水(6∶2.2∶1)]、点样量(1、2、3、4、5和6μL)、薄层板(硅胶G板、高效硅胶G板、硅胶GF254板、硅胶H板、聚酰胺薄膜)等薄层色谱条件进行考察。结果以正丁醇-冰乙酸-水(6∶2.2∶1)为展开系统,0.2%茚三酮为显色剂,硅胶G板为薄层板,混合氨基酸对照品溶液和供试品溶液的点样量分别是2μL和3μL,薄层层析法鉴别出桑枝中含有精氨酸,苏氨酸和缬氨酸,其薄层色谱图斑点清晰,专属性强。结论该方法简便可行,结果准确,可作为桑枝中氨基酸的薄层鉴别方法。ObjectiveTo establish the methods for identification of amino acids in Ramulus Mori by TLC.MethodsThe amino acids was extracted by ultrasonic method with distilled water as solvent,the TLC conditions,such as the development system[n-butanol-acetic acid-water(4∶2.2∶3),n-butanol-acetic acid-water(4∶1∶5),n-butanol-acetic acid-water(4∶3∶1),n-butanol-acetic acid-water(4∶2.2∶1),n-butanol-acetic acid-water(6∶2.2∶1)],spotting sample volume(1,2,3,4,5 and 6μL),thin layer chromatography plate(G plate,efficient silica gel G plate,GF254 plate,H plate,polyamide film),were investigated.ResultsThe TLC analysis indicated that the test samples contained arginine,threonine and valine.The spots were clear and its specificity was high when n-butanol-acetic acid-water(6∶2.2∶1)was used as the developing system,and 0.2%ninhydrin as chromogenic reagent,G plate was used as thin layer chromatography plate,and the spotting sample volume of amino acid mixed standard and sample were 2μL and 3μL respectively.ConclusionThe methods are simple,accurate and can be used as amino acids in Ramulus Mori identification.
