摘 要:目的:研究肿瘤相关中性粒细胞对人骨肉瘤细胞迁移能力的影响及可能的机制。方法:HE和免疫组化染色检测骨肉瘤临床标本中中性粒细胞浸润程度;中性粒细胞与人骨肉瘤细胞系SOSP-9607共培养,x CELLigence RTCA DP多功能实时无标记细胞分析仪监测不同比例中性粒细胞对SOSP-9607细胞迁移能力的影响;细胞因子芯片检测中性粒细胞与人骨肉瘤细胞系SOSP-9607共培养后细胞因子变化情况。结果:在临床转移骨肉瘤标本中HE和免疫组化染色均显示肿瘤组织内存在中性粒细胞的浸润;x CELLigence RTCA DP多功能实时无标记细胞分析仪监测显示,骨肉瘤细胞SOSP-9607和人外周血中性粒细胞共培养后细胞迁移能力明显增强于对照组;细胞因子芯片检测发现IL-6、IL-1β、Angiogenin、IGFBP-1等多个细胞因子明显增高。结论:人骨肉瘤细胞SOSP-9607和肿瘤相关中性粒细胞共培养,中性粒细胞能够促进SOSP-9607细胞的迁移,而这种促进作用很可能是通过IL-6、IL-1β、Angiogenin、IGFBP-1等多种细胞因子实现的。Objective:To investigate the effect of tumor associated neutrophils on the migration of osteosarcoma cells and its possible mechanism.Methods:The extent of neutrophil infiltration in osteosarcoma specimens was studied by HE and immunohistochemical staining.Neutrophils were cultured with human osteosarcoma cells SOSP-9607,and the migration of SOSP-9607 cells co-cultured with different proportions of neutrophils were monitored using xCELLigence RTCA DP.Cytokine microarray was performed to detect the changes of different cytokines after co-culture.Results:HE and immunohistochemical staining showed that there was infiltration of neutrophils in clinical osteosarcoma specimens.xCELLigence RTCA DP showed that the migration ability of SOSP-9607 cells was significantly enhanced when they were co-cultured with neutrophils.Cytokine microarray analysis showed that IL-6,IL-1β,Angiogenin,IGFBP-1 and other cytokines were significantly increased.Conclusion:Neutrophils can promote SOSP-9607 migration when co-cultured with SOSP-9607 cells,which is likely to be achieved via a variety of cytokines such as IL-6,IL-1β,Angiogenin and IGFBP-1.
