高静水压下益气活血汤通过p38MAPK信号通路对兔椎体终板软骨细胞的调控作用

摘　　要：目的观察益气活血汤含药血清在高静水压下对兔终板软骨细胞p38MAPK信号通路的调控作用,探讨益气活血汤治疗椎间盘退行性病变的可能作用机制。方法选用3月龄新西兰白兔制备益气活血汤含药血清,体外分离培养第3代兔椎体终板软骨细胞,建立体外高静水压加载干预模型,确定益气活血汤含药血清最佳干预条件。将体外培养的第3代终板软骨细胞随机分为空白组、模型组及含药血清组,空白组无任何干预,其余2组分别施加1 MPa静水压干预24 h后收集终板软骨细胞,运用Western Blot法检测各组终板软骨细胞p38、磷酸化p38蛋白表达情况。结果持续高静水压干预下,1μg/μL含药血清促终板软骨细胞增殖作用最明显。各组终板软骨细胞p38蛋白表达量比较差异无统计学意义;模型组磷酸化p38蛋白表达量显著高于空白组,而含药血清组磷酸化p38蛋白表达量显著低于模型组(P<0.05)。结论益气活血汤可以提高持续高静水压下终板软骨细胞增殖率,抑制软骨细胞凋亡,可能与其降低磷酸化p38蛋白表达水平,抑制p38MAPK信号通路的激活有关,推测益气活血汤可能通过调控p38MAPK信号通路发挥延缓椎间盘退行性病变的作用。Objective It is to observe the regulating effects of serum medicated with Yiqihuoxue decoction on the highhydrostatic pressure of endplate cartilage cell in vitro culture based upon p38MAPKsignal pathway and probe the mechanism of this decotion in the treatment of disc degeneration disease.Methods The preparation of Yiqihuoxue decoction containing medicine serum with 3 months old New Zealand rabbits,then endplate cartilage cell from 3 months old New Zealand were isolated and harvested in vitro.The model of endplate cartilage cell uploaded by the high hydrostatic pressure was established,the best intervention time of medicated serum was determined.The endplate cartilage cells of third generation in vitro were randomly divided into three groups:blank group,model group and drug containing serum group.The cells in the blank group was given no intervention,the cells in the other two groups were collected after uploading by the high hydrostatic pressure(1MPa)for 24 h.The Western Blot method was used to detect the expression of endplate cartilage cells p-p38 protein andp38 protein in every group.Results 1μg/uL drug containing serum showed the greatest effect on promoting the endplate cartilage cell proliferation under continuous high hydrostatic pressure intervened,.There was no statistical difference in p38 expression among the 3 groups(P>0.05).The expression of p-p38 protein in the model group was significantly higher than that in blank group,meanwhile,the expression in the drug containing serum group was significantly lower than that in model group(P<0.05).Conclusion Yiqihuoxue decoction can inhibitthe p38MAPK signal pathway,which can effectively promote the endplate cartilage cell proliferation.We speculated that Yiqihuoxue decoction may play a role of delaying intervertebral disc degenerative diseases through regulating p38MAPK signaling pathways.
