地龙粉酶解用酶初步筛选

摘　　要：分别利用胃蛋白酶、胰蛋白酶、胰凝乳蛋白酶、弹性蛋白酶、木瓜蛋白酶对地龙粉进行酶解,以水解度和对肺腺癌细胞抑制率为指标筛选最佳用酶。利用五种酶对地龙粉进行酶解,同时进行空白实验,采用MTT体外抗肿瘤技术,通过水解度和酶解液对肺腺癌细胞的抑制率筛选最佳用酶。胃蛋白酶和胰凝乳蛋白酶对地龙粉的水解度分别为9.42%、7.95%,且两者的酶解液对肺腺癌细胞A549的抑制率与浓度成负相关;木瓜蛋白酶对地龙粉的水解度最低且酶解液对肺腺癌细胞A549无抑制作用;胰蛋白酶对地龙粉的水解度为17.96%,其酶解液对肺腺癌细胞A549在低浓度下无抑制作用,在1 mg/L时抑制作用达最大,为83.08%;弹性蛋白酶对地龙粉的水解度最大,为30.61%,且其酶解液对肺腺癌细胞A549的抑制作用与浓度成正相关。地龙粉酶解最佳用酶为弹性蛋白酶和胰蛋白酶混合使用。The earthworm powders were enzymatically hydrolyzed by pepsin,trypsin,chymotrypsin,elastase,and papain.The best enzymes were selected by their hydrolysis degrees and inhibition rates of lung adenocarcinoma cells.Five kinds of enzymes were used in enzymatic hydrolysis of the earthworm powders.The best enzymes were selected by the hydrolysis degrees of enzymes and the inhibition rates of lung adenocarcinoma A549 cell.The hydrolysis degrees of earthworm powders using pepsin and chymotrypsin were 9.42%and 7.95%.respectively,the inhibitory rates of the two enzymes were negatively correlated with the concentration of adenocarcinoma cell A549;Papain has the lowest hydrolysis degree of the earthworm powders and the enzyme solution has no inhibitory effect on the lung adenocarcinoma cell A549;The hydrolysis degree of trypsin was 17.96%,the enzyme solution has no inhibitory effect on the low concentration of lung adenocarcinoma cell A549,the maximum inhibition is 83.08%at the concentration of 1mg/l;Elastase has the greatest hydrolysis rate of 30.61%,and the inhibition of the enzyme solution to the pulmonary adenocarcinoma A549 is directly related to its concentration.The best enzyme for the enzymatic hydrolysis of earthworm powders is the combination use of elastase and trypsin.
