摘 要:目的探讨云南产油菜蜂花粉多糖(rape bee pollen polysaccharide,RBPP)的肝脏保护作用。方法取50只小鼠随机分成RBPP高、中、低剂量组、模型组、空白对照组。各组给于相应的药物溶液10 ml/kg,正常对照组与肝损伤模型组给予等体积生理盐水,每天1次,连续给药14 d,末次给药2 h后模型组、多糖高、中、低剂量组腹腔注射0.1%CCl_4花生油溶液10 ml/kg,空白对照组腹腔注射等体积花生油溶液,禁食不禁水12 h后断头取血,离心取血清,剖腹取肝脏,分别检测血清天门冬氨酸氨基转氨酶(AST)、丙氨酸氨基转氨酶(ALT),肝组织超氧化物歧化酶(SOD)活性及丙二醛(MDA),观察肝脏组织学病理改变。结果与模型组相比,RBPP高、中、低剂量组均可显著降低血清ALT、肝组织MDA含量,升高肝组织SOD活性(P<0.01);PBPP高、中剂量组可显著降低血清AST含量(P<0.01),RBPP低剂量组血清AST无显著降低;小鼠肝组织的病理组织学检查结果表明,RBPP高、中剂量组均能使小鼠肝细胞的肿胀、坏死、充血等症状明显减轻,而RBPP低剂量组未见明显改善。结论 RBPP对急性肝损伤小鼠具有显著的肝脏保护作用。Objective To investigate the protective effects of Yunnan rape bee pollen polysaccharide(RBPP)on acute liver injury.Methods Fifty Mice were randomly divided into 5 groups:blank control group,model control group,RBPP high,middle and low dose groups.The animals in model control group and blank control group were given normal saline and other groups were given 10 ml/kg of corresponding polysaccharide solution by intragastric infusion once a day for 14 days.2 h after the last intragastric infusion,the mice in the model control group and RBPP high,middle and low dose groups were intraperitoneally injected 10 ml/kg of 0.1%CCl4 peanut oil solution and the blank control group was given the peanut oil solution.After abstaining from food except water for 12 h,all mice were sacrificed and the livers was taken to observe the histopathological changes.And the activities of serum AST,ALT and SOD in liver tissue and the levels of MDA in liver tissue were detected.Results Compared with the model group,the activities of serum ALT and the levels of MDA in liver tissue were significantly reduced,while the activities of SOD in liver tissue increased(P<0.01)in RBPP high,middle and low dose groups.The activities of serum AST in RBPP high and middle dose groups were reduced remarkably(P<0.01),while the activities of serum AST in RBPP low dose group showed no significant decrease.The results from liver histopathological examination showed that the symptoms of swelling,necrosis,hyperemia of liver cells in RBPP high and middle dose groups were significantly reduced,but the symptoms were not obviously improved in RBPP low dose group.Conclusion RBPP possesses obvious protective effects on mice with acute liver injury.
