摘  要:目的研究高脂饮食对雌性大鼠性发育相关基因下丘脑GnRH mRNA及垂体GnRH-R mRNA表达的影响。方法 20只21日龄的雌性SD大鼠按配对设计分组原则,分为高脂组、标准组,每组10只。高脂组大鼠饲喂高脂肪饲料,正常组大鼠饲喂标准饲料。当高脂组大鼠全体出现阴道口开放后,于当天处死所有实验大鼠,获取下丘脑及垂体。采用RT-PCR法,检测下丘脑GnRH mRNA及垂体GnRH-R mRNA的表达水平。结果高脂组下丘脑GnRH mRNA和垂体GnRH-R mRNA均高于标准组(均<0.05)。结论长期高脂饮食可致雌性大鼠HPGA功能提前启动,其机制可能与促进下丘脑GnRH mRNA和垂体GnRH-R m RN表达有关。Objective To research the influence of high fat diet on the female rats1 expression o f hypothalamus GnRH mRNA and pituitary GnRH-R mRNA.Methods Twenty female SD rats of 21 days old were randomly divided into the high-fat group and the control group,10 in each group.The High-fat group group was fed with high-fat feed and the control group was fed with standard feed.When the 10 rats in the high-fat group appeared to have vaginal opening,all the rats were going to be executed to obtain the hypothalami and pituitaries.The relative expression of hypothalamus GnRH mRNA and pituitary GnRH-R mRNA were detected by RT-PCR.Results The relative expression of hypothalamus GnRH mRNA and pituitary GnRH-R mRNA of high-fat group was much higher than that o f control group(P<0.05).Conclusions Long-term high-fat diet can cause the female rats'function o f HPGA to start ahead o f schedule,and the mechanism may be related to the promotion of hypothalamic GnRH mRNA and pituitary GnRH-R mRN expression.
