摘 要:目的探讨YPEL3在喉鳞状细胞癌中甲基化状况及其临床意义。方法应用实时荧光定量甲基化特异性聚合酶链反应(q MSP)分别检测91例LSCC患者癌组织及其配对的癌旁正常组织中YPEL3启动子甲基化水平,并分析其与临床病理特征之间的关系,构建受试者工作特征曲线()评估其诊断价值。结果喉鳞状细胞癌组织中YPEL3基因启动子甲基化率明显高于癌旁正常组织(<0.01),男性、有吸烟史、低分化的喉鳞状细胞癌患者的甲基化水平分别高于女性、无吸烟史及高中分化,差异均有统计学意义(均<0.05)。曲线下面积()高达0.697(95%=0.620~0.774,<0.01)。结论 YPEL3基因启动子异常甲基化与喉鳞状细胞癌的发生和发展有紧密的联系,可作为喉鳞状细胞癌患者诊断的潜在生物标记物。Objective To investigate the methylation level of YPEL3 in laryngeal squamous cell carcinoma(LSCC)and its clinical significance.MethodsThe methylation level of YPEL3promoter in 91 LSCC tissues and their paired normal tissues was measured by quantitative methylation-specific polymerase chain reaction(qMSP),and the relationship between YPEL3 methylation and clinicopathologic features was analyzed.A receiver operating characteristic(ROC)curve was created to assess diagnostic values.Results The methylation level of YPEL3 in LSCC was significantly higher than that in adjacent tissues(P<0.01).YPEL3 gene promoter methylation level also increased in male patients,and the patients with smoking history and poor difiFerentiation(allP<0.05).The area under the ROC curve(AUC)was up to 0.697(95%CI=0.620~0.774,P<0.01).Conclusions The aberrant methylation of YPEL3 promoter is closely related to the oc-currence and development of LSCC,which can be used as a potential biomarker for the diagnosis of LSCC.
