摘 要:目的:通过宏基因组学方法研究健康志愿者口腔微生物群落,为慢性牙周炎治疗转归及预后提供生物学基础。方法:提取健康志愿者唾液样本及龈上、龈下菌斑样本的总DNA,构建基因文库,测序并分析结果(进行物种注释和相对丰度计算),获得样本微生物群落。结果:测序结果显示健康志愿者口腔唾液样本可获得15个微生物属,龈上菌斑样本中可获得19个微生物属,龈下菌斑样本中可获得20个微生物属,微生物群落存在多样性。健康志愿者龈上微生物样本组、龈下微生物样本组及唾液样本组间微生物门、属的平均相对丰度比差异有统计学意义(P<0.05)。结论:结合第二代高通量测序技术的宏基因组学方法能够较好地获得健康志愿者口腔样本中的微生物群落,并初步分析各样本组中的微生物群落结构。The purpose of this study is to find out the oral microbial community in healthy volunteers by meta-genomic method,and to provide the biological basis for the prognosis and consequences of the treatment of chronic periodontitis.Methods:Total DNA was extracted from healthy volunteers saliva samples,supra-gingival and sub-gingival bacterial plaque samples,so as to built genomic libraries.After sequencing and analyzing the results,species were annotated,and their relative abundance was calculated too.Results:The sequencing results showed that there were 15 microbial genuses in saliva samples of healthy volunteers,and there were 20 microbial genuses in samples from healthy volunteers supra-gingival and sub-gingival plaques.The mean relative abundance ratio of each healthy volunteers microorganism sample group(saliva sample group,supra-gingival sample group and sub-gingival sample group)was significantly different(P<0.05).Conclusions:The meta-genomic method using the second generation high-throughput sequencing technique can obtain the microbial community in the oral samples of healthy volunteers well,and it is capable of helping analyze the microbial community structure in each sample group.
