摘  要:目的观察电针对面神经损伤兔面神经核中胶质细胞源性神经营养因子(GDNF)、胆碱乙酰转移酶(CHAT)表达及神经元细胞形态的影响,探讨电针对面神经损伤再生修复的作用机制。方法将76只健康家兔随机分为假手术组、模型组、电针组、西药组,其中假手术组4只,其余每组24只。除假手术组外,其余组均制备面神经损伤模型。术后假手术组、模型组不给予干预;西药组造模后第1天开始给予维生素B110 mg、维生素B_1250μg加地塞米松2 mg肌肉注射,1次/d,地塞米松每7 d减半,连续4周;电针组造模后第1天开始进行电针治疗,每天连续治疗30 min,治疗5 d后休息2 d,疗程4周。术后4周镜下观察面神经损伤后面神经核中神经元细胞形态,并分别于术后1,2,3,4周免疫组化法检测各组兔面神经核中GDNF、CHAT表达情况。结果与模型组比较,电针组、西药组神经细胞轮廓结构清晰完整,肿胀、空泡数目减少,细胞数目增多。术后2,3,4周,电针组、西药组GDNF阳性表达数均明显高于模型组(P均<0.05),且电针组明显高于西药组(P均<0.05)。术后1,2,3,4周,模型组CHAT阳性表达数均明显低于假手术组(P均<0.05),且呈先下降后上升的趋势;电针组、西药组CHAT阳性表达数随时间增加而增高,相邻两治疗阶段比较差异有统计学意义(P<0.05),且电针组各时间段CHAT阳性表达数均明显高于模型组和西药组(P均<0.05)。结论电针刺激有促进面神经损伤修复的作用,其机制可能与增加面神经核中GDNF、CHAT的表达有关。Objective:It is to observe the effect of electroacupuncture on the expression of glial cell line-derived neurotrophic factor(GDNF),choline acetyltransferase(CHAT)and the morphology of neurons in the facial nucleus of rabbits with facial nerve injury,and explore the mechanism of electroacupuncture on regenerative repair of facial nerve injury.Methods 0x09 76 healthy rabbits were randomly divided into sham operation group,model group,electroacupuncture group and western medicine group.Among them,there were 4 cases in the sham operation groups and 24 cases in each of the other groups.Except the sham operation group,the other groups were prepared for facial nerve injury model.After operation,the rats in the sham operation group and the model group were not given any intervention.After first days,the rats in the western medicine group were given vitamin B110 mg,vitamin B 12 50μg and dexamethasone 2 mg for intramuscular injection,1 times per day,dexamethasone was reduced to half per 7 d for 4 weeks.Electroacupuncture group was treated with electroacupuncture at first days after the electroacupuncture group.30 min was treated continuously every day.2 days for rest per treatment of 5 days,and the treatment course was 4 weeks.The morphology of neurons in the nucleus of the nerve after facial nerve injury was observed at 4 weeks after the operation.The expression of GDNF and CHAT in the nucleus of facial nerve in each group was detected at 1,2,3,and 4-week after operation.Results 0x09 Compared with the model group,the contour of the nerve cells in the electroacupuncture group and the western medicine group were clear and complete,the number of swelling and vacuoles were decreased,and the number of cells was increased.At 2,3,4-week after operation,the positive expression of GDNF in the electroacupuncture group and the western medicine group was significantly higher than that in the model group(all P<0.05),and the electroacupuncture group was significantly higher than that in the western medicine group(all P<0.05).The posit
