摘 要:为提高汉逊氏葡糖酸醋杆菌(Gluconacetobacter hasenii)JR-02细菌纤维素(BC)合成能力,采用响应面法对其发酵工艺参数进行优化。通过单因素试验和Plackett-Burman试验筛选出对菌株JR-02的细菌纤维素产量影响较为显著的4个因素为初始pH、Mg SO_4、玉米浆干粉、乙醇,再通过Box-Behnken试验设计最终确定最佳发酵工艺为:葡萄糖30 g/L、玉米浆干粉15 g/L、乙酸3 g/L、Na_2HPO_43 g/L、Mg SO_40.11 g/L、Ca CO_30.8 g/L、乙醇9 mL/L、初始p H 4.5、30℃、160 r/min。优化后BC产量达到(4.74±0.15)g/L,为未优化前BC产量的1.87倍。The fermentation process parameters were optimized by response surface method for improving the bacterial cellulose(BC)synthesis capacity of Gluconacetobacter hasenii JR-02.Four factors which had significant effect on bacterial cellulose yield were determined as initial pH,MgSO4,corn steep powder and ethanol by single factor and Plackett-Burman experiments.The optimum fermentation process was determined as glucose 30 g/L,corn steep powder 15 g/L,acetic acid 3 g/L,Na2HPO4 3 g/L,MgSO4 0.11 g/L,CaCO3 0.8 g/L,ethanol 9 ml/L,initial pH 4.5,culturing at 30 t and 160 r/min.After optimization,bacterial cellulose yield was up to(4.74±0.15)g/L,which was 1.87 times higher than that of before optimization.
