外加碳源对耐高温采油菌株ZY-3生长和代谢的影响

摘  要:分别以木糖、淀粉、葡萄糖、石蜡以及石蜡与糖的混合物为碳源,在60℃下静置培养7 d,通过测定菌体生长、发酵液的表面张力、乳化活性和碳源的残留量,考察耐高温采油菌株ZY-3利用上述碳源的生长以及与表面活性相关的代谢。结果表明,菌株ZY-3可利用上述碳源生长,并合成生物表面活性剂。石蜡与葡萄糖或淀粉的混合碳源不明显促进细胞生长,可较大幅度降低发酵液表面张力,其长时间的发酵液的乳化活性也大幅度提高。淀粉能明显减少石蜡的消耗。石蜡与木糖的混合碳源有利于细胞生长,但对减少石蜡消耗和提高发酵液表面活性无明显作用。Thermophile strain ZY-3 for enhancing oil recovery was cultivated at 60℃for 7 d in the media with xylose,starch,glucose,paraffin and the mixture of paraffin and saccarides as carbon sources.The growth and metabolism related with surface activity of the strain was evaluated by measuring the bacteria growth,surface tension,emulsifying activity and the residues of carbon sources.The results showed that the strain ZY-3 could utilize the tested carbon sources for the growth of cells and the production of biosurfactants.The mixed carbon source of paraffin with glucose or starch could not obviously promote cell growth,but significantly reduce surface tension of fermented broth,the emulsification activity of the broth fermented for long time could also increased significantly.Starch could significantly reduce the consumption of paraffin.The mixed carbon source of paraffin and xylose could improve the growth of cells,but had no obvious effect on the reduction of paraffin consumption and the enhancement of surface activity of fermented broth.
