摘 要:以榆树1~2a生枝条为研究材料,以榆树总黄酮粗提物得率为指标,在单因素试验基础上,通过响应面法优化榆树总黄酮闪式提取工艺,并比较不同采收季节榆树1~2a生枝条中的总黄酮含量,结果表明:榆树总黄酮提取最佳工艺条件为浸泡温度40℃、料液比1∶60(g/mL)、乙醇浓度50%、闪提时间1min和闪提次数4次;不同采收季节榆树总黄酮含量存在差异,由大到小依次为夏季、秋季、春季;在上述最佳提取条件下以夏季1~2a生枝条为材料进行提取,黄酮粗提物得率达8.39%。Using 1 to 2 year-old Ulmus pumila branches as research materials and taking the yield of total flavonoids of crude extract from Ulmus pumila as the index,based on the single factor experiment,the response surface methodology(RSM)was employed to optimize the flash-extraction technology of total flavonoids in Ulmus pumila,and the contents of total flavonoids in the branches of 1 to 2 year-old Ulmus pumila were compared.The results showed that the optimum extraction conditions of total flavonoids from Ulmus pumila were as follows:soaking temperature was 40℃,ratio of material to liquid was 1∶60(g/mL),concentration of ethanol was 50%,flash-extraction time was 1 min and 4 times of flash-extraction.The content of total flavonoids was different in different harvest seasons,and the sequence in a decreasing order was summer,autumn and spring.Under the optimum extraction conditions,the crude extraction yield of total flavonoids from 1 to 2 year-old Ulmus pumila branches in summer was 8.39%.
