摘 要:目的通过建立BALB/c小鼠结直肠癌移植瘤模型,探讨植酸及其不同水解率产物对肿瘤细胞分化相关因子的影响。方法通过皮下注射方法建立结直肠癌移植瘤模型,将28只造模成功的小鼠按照肿瘤结节大小随机分为生理盐水组(A组)、30%植酸水解率组(B组)、90%植酸水解率组(C组)、植酸组(D组)4组,每组7只。4组采用瘤内注射法分别注射生理盐水及浓度为100mg/L的30%植酸水解率产物、90%植酸水解率产物和植酸,每周2次。10周后处死小鼠,观察各组小鼠瘤体大小;采用Western blot方法检测肿瘤组织内来源于神经细胞瘤的骨髓细胞瘤病毒相关癌基因(N-MYC)、N-MYC下游调节基因1(NDRG1)、同源磷酸酶-张力蛋白基因(PTEN)蛋白的表达。结果与A组相比,C组和D组小鼠肿瘤体积较小(P<0.05);NDRG1和PTEN蛋白表达水平升高,N-MYC蛋白表达水平降低(P<0.05);且C组的效果优于D组(P<0.05)。结论植酸及其水解产物可能通过上调NDRG1蛋白的表达来诱导肿瘤分化和抑制肿瘤生长,且90%植酸水解率产物的效果优于植酸。Objective To investigate the effect of phytic acid and its hydrolysates with different hydrolysis rates on tumor cell differentiation-related factors by establishing a xenograft tumor model of colorectal cancer in BALB/c mice.Methods Subcutaneous injection was performed to establish a xenograt tumor model of colorectal cancer.According to the size of tumor nodules,28 mice after modeling were randomly divided into normal saline group(group A).30%hydrolys is rate group(group B),90%hydrolysis rate group(groupC),and phytic acid group(group D),with 7 mice in each group.The mice in these four groups were given intratumoral injection of normal saline,100 mg/L hydrolysate with ahydrolysis rateof 30%or 90%,or phytic acid twice aweek.The mice were sacrificed 10 weeks later,and the size of tumor was observed:Western blot was used to measure the protein expression of myelocytomatosis viral related oncogene,neuroblastoma derived(N-MYC),N-MYC downstream-regulated gene 1(NDRG1),and phosphatase and tensinhomolog(PTEN)in tumor tissue.Results Compared with group A,groupsC and D had a significantly smaller tumor size(P<0.05),significant increases in the protein expression of NDRG1 and PTEN(P〈0.05),and a significant reduction in the protein expression of N-MYC(P〈0.05),and group C had significantly better results than group D(P〈0.05).Conclusion Phytic acid and its hydrolysates can induce tumor differentiation and inhibit tumor growth pos-sibly by upregulating the expression of NDRG1,and the hydrolysatewith a hydrolysis rate of 90%has a better effect than phytic acid.
