摘  要:目的观察骨髓间充质干细胞(BMSCs)与硫酸软骨素酶ABC(ChABC)联合移植对大鼠脊髓损伤的修复作用,并探讨其机制。方法 80只脊髓亚急性损伤大鼠随机分为4组各20只。损伤后第6天,A组先后移植2.0×10~6/μL的原代BMSCs细胞悬液、1 U/mL的ChABC各20μL,B组予等量BMSCs,C组予等量ChABC,D组予等量PBS。比较各组运动功能评分(BBB评分)、脊髓损伤区面积及后肢体感诱发电位(SEP)、经颅磁刺激运动诱发电位(MEP)潜伏期和胶质纤维酸性蛋白(GFAP)、生长相关蛋白-43(GAP-43)、脑源性神经生长因子(BDNF)阳性细胞百分比。结果与D组比较,其余3组制模后第1天BBB评分降低,第14、28天BBB评分升高;与A组比较,B、C组BBB评分降低(P均<0.05)。与同组移植前比较,其余3组SEP、MEP潜伏期短;与A组比较,B组SEP潜伏期长,C、D组SEP潜伏期短;与A组比较,其余3组MEPP潜伏期短;P均<0.05。与其余3组比较,A组损伤区面积小(P均<0.05)。与D组比较,其余3组GFAP阳性细胞百分比低,GAP-43、BDNF阳性细胞百分比高;与A组比较,GFAP阳性细胞百分比高,GAP-43、BDNF阳性细胞百分比低;P均<0.05。结论 BMSCs与ChABC联合移植对大鼠脊髓损伤有修复作用,机制可能与其降低GFAP、GAP-43表达及升高BDNF表达有关。Objective To explore the role and related mechanism of combined transplantation of chondroitinase ABC(ChABC)and bone marrow stromal cells(BMSCs)during the repair process of rats with spinal cord injury.Methods Eighty SD rats were randomly divided into four groups(A,B,C,and D)with 20 rats in each group.On the sixth day after injury,the rats in group A were transplanted with 20μL of 2.0×10 6/μL original BMSCs suspension and 1 U/mL ChABC,while the same dose of BMSCs,ChABC,and PBS were transplanted into group B,C,and D respectively.The BBB score,spinal cord injury area,somatosensory evoked potential(SEP),motor evoked potential(MEP)latency,and the percentage of cells with positive expression of glial fibrillary acidic protein(GFAP),growth-associated protein 43(GAP 43),and brain-derived neurotrophic factor(BDNF)were compared among different groups.Results Compared with group D,BBB scores of the other three groups were lower on day 1 after transplantation,and became higher on the 14th and 28th day after transplantation.Compared with group A,BBB scores of group B and C were lower(both P<0.05).Compared with the same group before treatment,the latency of SEP and MEP in the groups A,B and C was short;compared with group A,the latency of SEP in the group B was longer while shorter in the groups C and D;compared with group A,the latency in the other three groups were shorter(all P<0.05).The injury area was the smallest in the group A as compared with that of the other three groups(all P<0.05).Compared with group D,the percentage of GFAP positive cells was lower while the percentage of GAP-43 and BNDF positive cells was higher in the other three groups.Compared with group A,the percentage of GFAP positive cells was higher while the percentage of GAP-43 and BNDF positive cells was lower in the other three groups(all P<0.05).Conclusion Transplantation of ChABC combined with BMSCs can help repair the spinal cord injury in rats,and the mechanism may be related to the down-regulation of the GFAP,GAP-43 and the up-regulation of
