摘 要:目的:检测姜黄素对肝癌SMMC-7721细胞增殖及凋亡的影响,并进一步探讨内质网应激反应在其中的作用。方法:应用2.5μM、5μM、10μM、20μM、50μM的姜黄素处理肝癌SMMC-7721细胞,采用四甲基偶氮唑蓝(MTT)法检测细胞活性;Western Blot检测内质网应激相关蛋白GRP78、caspase-12、CHOP的表达;流式细胞术检测姜黄素对肝癌细胞凋亡的影响。结果:与对照组比较,2.5μM、5μM、10μM、20μM、50μM的姜黄素对肝癌SMMC-7721细胞增殖均有抑制作用,并随作用时间的增加,增殖抑制作用更加显著(均P<0.05);姜黄素能够诱导肝癌SMMC-7721细胞GRP78(P<0.05)、caspase-12(P<0.05)、CHOP(P<0.01)的表达,其表达量具有剂量依赖性;姜黄素能够诱导肝癌SMMC-7721细胞凋亡,与对照组比较(P<0.05),呈现出剂量依赖关系。结论:姜黄素通过内质网应激信号通路诱导肝癌细胞凋亡,抑制其增殖。Objective:To investigate the effect of curcumin on ERS-related apoptosis and cell proliferation in smmc-7721 cells.Methods:SMMC-7721 cells were incubated with the blank vehicle,2.5,5,10,20μM of curcumin.After intervention,cell proliferation were detected by MTT,protein expressions of GRP78,caspase-12,CHOP were assessed by western blotting.Cell apoptotic rate were detected by flow cytometry.Results:Compared with the control group,2.5,5,10,20μM cucurmin inhibited the proliferation of hepatocellular carcinoma SMMC-7721 cells,and with the increase of the time,the inhibition of proliferation is more significant(All P<0.05).Curcumin could induce the expression of GRP78(P<0.05),caspase-12(P<0.05)and CHOP(P<0.01)in SMMC-7721 cells,and the expression of these proteins structures were dose dependent.Curcumin induced apoptosis of hepatocellular carcinoma SMMC-7721 cells,compared with the control group(P<0.05),showing a dose-dependent relationship.Conclusion:Curcumin may suppresse cell proliferation and induce apoptosis partly through activation of ERS signaling pathway in hepatocellular carcinoma cells.
