摘  要:双歧杆菌是人和动物肠道中一种重要的益生菌,具有许多生理和保健功能。实验使用厌氧培养箱培养筛选获得双歧杆菌;经鉴定为革兰氏阳性菌(G+)。选用海藻酸钠作为成膜材料、羧甲基甲壳素作为保护剂、Ca Cl_2作为固化液,通过挤压法将双歧杆菌包埋在了微胶囊里;将得到的微胶囊分别置于胃液、肠液中并测定其溶解情况,得到了制备微胶囊的配比和最适浓度。使用2.14%海藻酸钠、1.79%羧甲基甲壳素制备双歧杆菌微胶囊,能够得到最好的包埋效果,菌体包埋率可达66.81%;此结果可为双歧杆菌微胶囊的高效生产提供一个参考。Bifidobacterium is an important probiotics in intestines of human and animals,which plays crucial roles in a lot of physiological and healthy function.Bifidobacterium bacteria samples were cultivated and screened in anaerobic incubator.Selected Bifidobacterium showed bluish violet of Gram-positive bacteria(G+)color under microscope with Gram staining method.The optimal ratio of involved materials was explored for Bifidobacteria microcapsules which could go through gastric juice and release in intestine.An extrusion method was used to prepare Bifidobacteria microcapsules,which involved sodium alginate as film-forming material,carboxymethyl chitin as a protective agent and CaCl 2 as curing liquid.Optimal ratio of related materials was obtained in preparing microcapsules by determining dissolutions of immersing resulted microcapsule in gastric juice and intestinal juice respectively.The embedding rate reached 66.81%when using 2.14%sodium alginate and 1.79%carboxymethyl chitin in the preparation of Bifidobacteria microcapsules.It can be a useful reference to produce Bifidobacteria microcapsules with high efficiency.
