摘 要:目的 探讨miR-489对黑色素瘤迁移的抑制作用。方法 通过Transwell实验检测miR-489对黑色素瘤迁移的影响;荧光素酶报告基因实验检测A375细胞中miR-489对PAK5的靶向作用;进一步通过Western blotting以及实时PCR检测miR-489对于A375细胞中PAK5的影响。结果 Transwell实验发现miR-489过表达后可以显著抑制A375细胞的迁移,当miR-489受到抑制后,A375细胞的迁移受到促进。荧光素酶报告基因实验证明,miR-489可以直接作用于PAK5。Western blotting和实时PCR检测发现,过表达miR-489可以抑制A375细胞中PAK5蛋白的表达;当miR-489受到抑制后蛋白表达有所增加。结论 miR-489可以通过打靶PAK5抑制黑色素瘤细胞的迁移。Objective To investigate the role of miR-489 in inhibiting the migration of melanoma cells.Methods The effect of miR-489 on the migration of melanoma cells was detected using the Transwell assay.Luciferase reporter gene assay was used to detect the effect of miR-489 on PAK5 in A375 human melanoma cells.Western blotting and real-time PCR were performed to test the effect of miR-489 on PAK5 in A375 cells.Results The Transwell assay showed that overexpression of miR-489 could significantly inhibit the migration of A375 cells,and that the migration of A375 cells was promoted when miR-489 was inhibited.Luciferase reporter gene experiments demonstrated that miR-489 could act directly on PAK5.Western blotting and real time PCR assay showed that overexpression of miR-489 could inhibit the expression of PAK5 protein in A375 cells,and the expression of PAK5 was increased when miR-489 was inhibited.Conclusion miR-489 can inhibit the migration of melanoma cells by targeting PAK5.
