沉默大鼠颌下腺细胞SSB1对NBS1蛋白表达的影响

摘　　要：目的:初步研究沉默大鼠单链DNA结合蛋白1(SSB1)基因对NBS1蛋白表达的影响。方法:将大鼠颌下腺(SMG)细胞分为病毒转染组、空白质粒对照组,分别转染rAdE5-SSB1-1p2-shRNA腺病毒表达载体、空载腺病毒,另设正常组,通过western blotting法检验SSB1、NBS1蛋白表达量。结果:病毒转染组NBS1、SSB1蛋白相对表达量均较空白质粒对照组、正常组显著降低,差异均有统计学意义(均P<0.05)。结论:成功沉默大鼠SMG细胞SSB1基因,SSB1沉默可下调NBS1蛋白表达。Objective:To investigate the protein expression of NBS1 after single-stranded DNA-binding pro-tein 1(SSB1)gene silencing.Methods:The submandibular gland(SMG)cells of rats were cultured and ran-domly divided into 3 groups:the virus group in which the cells were transfected with rAdE5-SSBl-1 p2-shR-NA adenovirus expression vector,the blank control group in which the cells were transfected with empty adenovirus?and normal control group.The protein expressions of SSB1 and NBS1 were detected by western blotting.Results:The expressions of NBS1 and SSB1 in the virus group were significantly lower than those in the blank control group and normal control group(P<0.05).(Conclusion:SSB1 gene was successfully si-lenced in the SMG cells of rats.SSB1 silencing could down-regulate the protein expression of NBS1
