摘 要:木糖渣含有大量的纤维素,可以用作生产燃料乙醇的原料。以木糖渣为原料,研究了酶用量及底物浓度对于乙醇浓度的影响。结果表明,当底物浓度为10 g/100 m L,酶用量由15 FPU/g底物增加到35 FPU/g底物,发酵72 h乙醇浓度由22.5 g/L增加到32.0 g/L。对发酵时间为48 h时酶用量与乙醇浓度进行了拟合,拟合后方程的相关系数R^2为0.955;当底物浓度由5 g/100 m L增加到20 g/100 m L时,发酵96 h时乙醇浓度分别为12.1 g/L、29.0 g/L、38.5 g/L、37.5 g/L。采用补料同步糖化发酵,进一步研究了补料次数对于乙醇浓度的影响,结果表明,初始底物浓度为15 g/100 m L,经过1~5次补料,补料后的总底物浓度为30 g/100 m L,发酵120 h补料次数为2次的乙醇浓度可以达到70.0 g/L,方差分析表明,补料次数对于乙醇浓度没有显著影响。对比了不同补料量对于乙醇浓度的影响,结果表明,初始底物浓度为15 g/100 m L,补料至底物浓度分别为40 g/100 m L、50 g/100 m L、52.5 g/100 m L时,发酵96 h乙醇浓度分别为65.2 g/L、62.1g/L、63.5 g/L。研究了半同步糖化发酵对于乙醇浓度的影响,发酵48 h乙醇浓度仅为14.4 g/L。Corncob residues(CCR)contain large amount of cellulose and it could be used as raw materials to produce fuel ethanol.In this study,the effects of the use level of cellulase and CCR concentration on ethanol production were explored.The results showed that,the concentration of ethanol increased from 22.5 g/L to 32.0 g/L after 72 h of SSF when CCR concentration was 10.0 g/100 mL and cellulase use level increased from 15 FPU/g substrate to 35 FPU/g substrate.A polynomial fitting of cellulase use level and ethanol concentration was performed at 48 h after the fermentation and the coefficient determination(R2)of the model was 0.955.As CCR concentration increased from 5 g/100 mL to 20 g/100 mL,ethanol concentration were 12.1 g/L,29.0 g/L,38.5 g/L,and 37.5 g/L respectively after 96 h fermentation.Then fed batch fermentation was performed to explore the effects of material feeding times on ethanol concentration,The results showed that,as CCR concentration was 15 g/100 mL,after 1 to 5 times material feeding respectively,CCR concentration was 30 g/100mL,the maximum ethanol concentration reached up to 70.0 g/L after 120 h fed batch SSF,and there was no significant difference in ethanol concentration for different fed batch.Besides,the effects of fed batch volume on ethanol concentration were studied.The results suggested that,ethanol concentration were 65.2 g/L,62.1 g/L,63.5 g/L after 96 h of fed batch SSF,as the initial substrate concentration was 15.0 g/100 mL,then to the end substrate concentration was 40.0 g/100 mL,50.0 g/100 mL,52.5 g/100 mL respectively by fed batch.A half SSF was performed.The results showed that ethanol concentration was 14.4 g/L after 48 h of fermentation,which implied that half SSF had a poor performance compared with SSF.
